摘 要:研究了8种莲子心水提物(LPWE)的抗炎活性。采用反相高效液相色潽法(RP-HPLC)分析8种莲子心水提物中黄酮类化合物,结果夏佛碳苷(含量为0.97%~1.47%)是所有水提物中含量最高的黄酮类化合物。以脂多糖(LPS)刺激小鼠腹腔巨噬细胞(RAW264.7)建立炎症模型,评价莲子心水提物和夏佛塔苷的抗炎活性,分析其含量与抗炎活性的相关性。研究发现40μg/m L和80μg/m L莲子心水提物可显著降低炎性介质NO的释放量和炎性因子TNF-α的分泌量,且对NO释放量的抑制作用呈现剂量依赖性。当莲子心水提物含量为80μg/m L时,其对NO的抑制作用与其含有的夏佛塔苷量呈显著正相关(R2=0.974;P<0.05)。研究表明8种莲子心水提物均具有抗炎作用,夏佛塔苷是其关键抗炎活性成分。选择夏佛塔苷含量较高的莲子心,可获得更好的抗炎效果。The anti-inflammatory activity of water extract from 8 lotus plumule were studied in this paper. Firstly,the composition of flavones in water extract from lotus plumule(LPWE) was analysed by reversed-phase high performance liquid chromatogram( RP-HPLC). And schaftoside was the highest content with a percentage from 0. 97 %to 1.47% among all the flavones in the LPWEs. Secondly, RAW264.7 macrophages was stimulated by lipopolysaccharide to make an inflammatory model. And the anti-inflammatory activity of LPWE as well as schaftoside was evaluated. The correlation analysis between dose and activity was followed. The results showed that LPWE significantly reduced the expression of NO and the level of TNF-α. And the higher concentration of LPWE, the better anti-inflammatory activity was. What's more, the NO inhibition rate of 80 μg/m L LPWE was significantly(P〈0.05) positive related with the content of schaftoside contained in it(R^2= 0.974). The research indicated that LPWE has a good anti-inflammatory activity, and schaftoside might be the key composition of LPWE's anti-inflammatory activity. Hence, choosing lotus plumule with a higher content of schaftoside might get a better anti-inflammatory activity.
