摘 要:目的建立白术中3种内酯成分的一测多评方法。方法采用超高效液相色谱法,以乙腈-水为流动相进行梯度洗脱,检测波长为235 nm,进样量为2μL,流速为0.2 mL·min^(-1),柱温30℃。以白术内酯Ⅲ为内参物,测定其与白术内酯Ⅰ、Ⅱ的相对校正因子,利用该相对校正因子计算其他2种成分的含量,同时利用外标法测定3种内酯的含量,比较2种结果的差异,验证一测多评法的可行性和准确性。结果在线性范围内,白术内酯Ⅲ与白术内酯Ⅰ、Ⅱ的相对校正因子分别为0.633、1.895,在不同实验条件下重现性良好,3种成分含量计算值与实测值间无显著性差异。结论所建立的一测多评法可用于白术中3种内酯成分的含量测定,可为白术的质量控制提供科学的理论依据。OBJECTIVE To establish a quantitative analysis of multi-components by single marker(QAMS) for the determination of three atractylenolides in Atractylodes macrocephala Koidz. METHODS UPLC method was applied to determine atractylenolide I , Ⅱ and Ⅲ by external standard method (ESM)first. With this established UPLC method, atractylenolide Ⅲ was used as the inter- nal standard (IS)to determine the correction factors (RCFs)of the two other atractylenolides, and their contents in all the samples were calculated by their RCFs at the same time. By comparing the contents determined by the ESM and QAMS methods, the feasibility and accuracy of QAMS method were verified. RESULTS Within a certain range ( atractylenolide I : 3.7 - 59.3 mL·min-1, atracty- lenolide Ⅱ : 3.9 -63.5 mL·min-1 , atractylenolide Ⅲ : 7.3 - 116.1 mL·min-1 ) , the RCFs of atractylenolide I , Ⅱ to at- ractylenolide Ill were 0. 633 and 1. 895, respectively, which had good repeatability in different experimental conditions. There was no significant difference between the QAMS method and ESM method(P 〉 0.05). CONCLUSION The QAMS method is feasible and accurate for the determination of the three atractylenolides and is beneficial to the quality control of Atractylodes mac- rocephala Koidz..
