摘 要:目的辐射通过产生自由基,诱导脂质过氧化介导组织细胞损伤,因此筛选高效低毒的辐射防护剂尤为迫切。本研究探讨山楂水提取物对辐射损伤小鼠生殖遗传系统的保护作用。方法选取健康SPF级昆明种雄性小鼠40只,随机分为对照组、模型组、山楂水提物高、中、低剂量组。对照组和模型组给予0.9%生理盐水,山楂水提物高、中、低剂量组给予不同剂量的山楂水提物1g/(kg·d)、0.5g/(kg·d)和0.25g/(kg·d)灌胃连续14d后接受4Gy的X射线一次性全身照射,24h后处死小鼠,计数小鼠骨髓细胞微核率,检测小鼠精子畸形率(Sperm deformity)及肝脏组织丙二醛(malondialdehyde,MDA)含量和超氧化物岐化酶(superoxide dismutase,SOD)活性。结果小鼠骨髓噬多染红细胞微核率结果显示,模型组微核率(24.10±2.92)‰明显高于对照组(3.54±0.25)‰,t=5.116,P<0.001,山楂水提物高剂量组(11.89±0.84)‰(P<0.001)、中剂量组(15.46±0.76)‰(P<0.001)、低剂量组(19.03±0.92)‰(P<0.001)小鼠骨髓微核数均低于模型组,F=223.74,P<0.001;小鼠精子畸形率结果显示,模型组精子畸形率(38.40±3.52)%明显高于对照组(3.30±0.26)%,t=4.561,P<0.001,山楂水提物高剂量组(11.35±1.72)%(P<0.001)、中剂量组(15.92±2.60)%(P<0.001)、低剂量组(20.40±3.07)%(P<0.001)小鼠精子畸形率均低于模型组,F=216.788,P<0.001;小鼠肝脏MDA含量结果显示,模型组肝脏MDA含量6.11±0.27,显著高于对照组1.65±0.12,t=6.711,P<0.001,与模型组相比,山楂水提物高剂量组2.43±0.22(P<0.001)、中剂量组3.11±0.20(P<0.001)、低剂量组4.30±0.32(P<0.001)MDA含量显著降低,F=439.752,P<0.001;SOD活性结果显示,模型组SOD活性33.84±2.33较对照组73.02±2.51显著下降,t=10.494,P<0.001,与模型组相比,山楂水提物高剂量组68.40±3.06(P<0.001)、中剂量组61.10±3.56,(P<0.001)、低剂量组51.68±3.31(P<0.001)SOD活性明显升高,F=216.245,P<0.001。结论山楂水提取物对X射线引起的急性辐OBJECTIVE By producing free radicals and lipid peroxidation,radiation can induce tissue and cell damage,thus screening efficient and low-toxicity radiation protection agents are particularly urgent.This study investigated the protective effects of Aqueous Extract of Crataegus on the agenetic reproductive system in mice with radiation injury.METHODS KM male mice(n=40)were randomly divided into control group,model group and Aqueous Extract of Crataegus high-dose,middle-dose and low-dose group.Mice in control group and model group were given 0.9% N.S,while mice in Aqueous Extract of Crataegus high-dose,middle-dose and low-dose group were given Aqueous Extract of Crataegus 1 g/(kg·d),0.5 g/(kg·d)and 0.25 g/(kg·d)by intragastric administration,once a day for 14 days,all groups except for control group were exposed to X-ray at the 15 th day.After 24 hirradiation,the bone marrow cell micronucleus rate,sperm malformation rate,malondialdehyde(MDA)content and activity of superoxide dismutase(SOD)in liver were measured.RESULTS Bone marrow cell micronucleus rate in model group(24.10±2.92)‰ was significantly increased compared with control group(3.54±0.25)‰(t=5.116,P〈0.001),compared with model group,the bone marrow cell micronucleus rates in Aqueous Extract of Crataegus high-dose(11.89±0.84)‰(P=0.000),middle-dose(15.46±0.76)‰(P=0.000)and low-dose group(19.03±0.92)‰(P〈0.001)were significantly decreased,F=223.74,P〈0.001.The sperm malformation rate in model group(38.40±3.52)% was significantly increased compared with control group(3.30±0.26)%(t=4.561,P〈0.001),the sperm malformation rate in Aqueous Extract of Crataegus high-dose(11.35±1.72)%(P〈0.001),middle-dose(15.92±2.60)%(P〈0.001)and low-dose group(20.40±3.07)%(P〈0.001)were remarkly decreased,F=216.788,P〈0.001.Also the MDA content in model group(6.11±0.27)was increased compared with control group(1.65±0.12),t=6.711,P〈0.001,while the
