摘 要:目的分析幽门螺杆菌(H.pylori)在胃息肉患者中的感染情况,利用高通量测序技术分析胃息肉患者的胃液菌群组成,探究整体菌群变化与息肉发生的关系。方法收集7例胃息肉患者的胃液(GP组),7例胃体黏膜未见异常体检者胃液为对照组(C组),统计H.pylori感染情况。提取细菌总DNA,采用高通量测序技术对16S rRNA基因的V3-V4高变区测序,分析比较菌群结构。结果 (1)7例胃息肉患者中5例H.pylori阳性,H.pylori感染率为71.4%;对照组与胃息肉组H.pylori感染率无差异。(2)两组之间菌群α多样性差异无统计学意义,β多样性有显著区别。门水平上,两组之间菌群差异无统计学意义;属水平上,胃息肉组胃液奈瑟菌属(P<0.05)、嗜血杆菌属(P<0.05)、Parvimonas属(P<0.05)比例显著增加。结论胃息肉患者胃液菌群发生紊乱,以奈瑟菌属、嗜血杆菌属、Parvimonas属显著增加为特征。Objective To detect Helicobacter pylori infection in patients with gastric polyps (Gps), and ana- lyze the changes in microflora in the gastric juice in patients with gastric polyps based on the analysis of high throughput sequencing technology and investigate the relationship between the incidence of polyps and the al- ternation of mieroflora. Methods We analyzed the H. pylori infection of 7 samples in the gastric juice in patients with gastric polyps (GP), and 7 samples in the gastric juice in physical examination people as con- trol (C), whose gastric body mucosa presented normal. Total genomic DNA was extracted. PCR amplifica- tion of V3 - V4 regions of bacterial 16S rRNA analyzed the microbial community structure by using the high throughput sequencing technology. Results (1) In group GP, 5 individuals with a clinical and histopatho- logical diagnosis of H. pylori-positive, the H. pylori infection ratio was 71.4%, and no difference were found between group C and group GP. (2) There was no difference in alpha diversity between group C and group GP, however there was significant difference between two groups in beta diversity. There was no clear difference between group C and group GP at phyla level, while at genus level, Neisseria, Haemophilus and Parvimonas increased remarkedly in group GP (P〈0.05) . Conclusion Ecological imbalance in the gastric juice flora in the patients with gastric polyps characterized by significant increasing of Neisseria, Haemophilus and Parvirnonas.
