摘  要:目的观察中药黄连低温提取物对体外培养的人恶性疟原虫的抑制和杀灭作用。方法采用低温冷冻离心法提取黄连有效成分,获得3个提取物。根据体外培养的恶性疟原虫在药物作用下其生长发育会受到抑制,甚至被杀灭的原理,将3个黄连提取物分别用MPI1640不完全培养基溶解,稀释至所需浓度,制作测定板。采用Rieckmann体外微量测定法,测定3个黄连提取物分别对疟原虫的作用,计算疟原虫在各浓度的抑制率。用ICEstimator software软件计算各种提取物的半数抑制浓度和完全抑制浓度。结果3个黄连提取物对恶性疟原虫均有一定的抑制和杀灭作用,从0.8 mg/L浓度井开始出现疟原抑制;提取物浓度为0.8、1.6、3.2、6.4 mg/L时,疟原虫在1号提取物的抑制率分别为1.3%、59.1%、94.0%和95.9%,在2号提取物抑制率分别为14.7%、40.2%、70.3%和95.9%,在3号提取物抑制率依次为3.4%、16.5%、89.5%和97.4%。1号提取物的IC_(50)、IC_(90)和IC_(99)分别为38.18、47.56、60.45 mg/L,2号提取物的IC_(50)、IC_(90)和IC_(99)分别为50.00、93.15、183.69 mg/L,3号提取物IC_(50)、IC_(90)和IC_(99)依次为51.30、87.40、156.08 mg/L。结论低温提取黄连有效成份的方法,能提取黄连中有效成份;中药黄连的这3个提取物具有杀灭疟原虫的作用,值得进一步研究。关键词:黄连;低温提取物;疟原虫;Objective To study the inhibiting and killing efficacy of Coptis chinensis extractives against Plasmodium falciparum in vitro. Methods Three extractives were obtained by low temperature freezing centrifugation. P. falciparum was cultured in vitro. The extractives were dissolved in MPI1640 incomplete medium and diluted to the required concentration and then added in the cells of culture plates. The inhibiting and killing efficacy of C. chinensis extractives against P. falciparum were tested by the Rieckmann in vitro micro test. The median inhibitory and complete inhibitory concentrations were calculated by using ICEstimator software for each extractive. Results The three extractives inhibited and killed P. falciparum at a certain degree. The dosage of 0.8 mg/L began to present the inhibition effect. The inhibition rates were 1.3%, 59.1%, 94.0% and 95.9% in 0.8, 1.6, 3.2 mg/L and 6.4 mg/L of C. chinensis extractive No.1 respectively; were 14.7%, 40.2%, 70.3% and 95.9% in above-mentioned concentrations of extractive No.2 respectively; were 3.4%, 16.5%, 89.5% and 97.4% in above- mentioned concentrations of extractive No. 3, respectively. IC50, IC90 and 1C99 of extractive No. 1 were 38.18 mg/L, 47.56 mg/L and 60.45 mg/L respectively; those of extractive No. 2 were 50.00 mg/L, 93.15 mg/L and 183.69 mg/L respectively; those of extractive No. 3 were 51.30, 87.40 mg/L and 156.08 mg/L respectively. Conclusion The method of low temperature freezing centrifugation can extract active ingredients from C. chinensis. These C. chinensis ingredients have plasmodicidal effect at some degree, and it deserves to do further researches.
