摘  要:目的:观察针刺对糖尿病抑郁模型大鼠血清CORT昼夜节律及SCN内Per1、Per2基因表达的影响,探讨针刺治疗糖尿病抑郁的生物学机制。方法:将36只SD大鼠随机分为空白组(12只)、模型组(12只)和针刺组(12只)3组。模型组和针刺组运用腹腔注射链脲佐菌素+慢性应激法建立糖尿病抑郁复合模型。针刺组于造模后每日取穴"后三里"(双)、"三阴交"(双)、"百会"针刺,每次留针20 min,每日1次,连续针刺7 d。空白组和模型组相同方法抓取固定,不予处理。针刺结束后检测各组血糖、旷场实验行为学变化、血清CORT昼夜节律及SCN内Per1、Per2基因的表达水平。结果:模型组血糖较空白组显著升高(P〈0.05),而针刺组大鼠血糖较模型组显著下降(P〈0.05);模型组水平跨格数和直立次数较空白组显著减少(P〈0.05),针刺组大鼠水平跨格数和直立次数较模型组显著增加(P〈0.05);模型组大鼠CORT较空白组显著升高(P〈0.05)且近似昼夜节律消失(P〉0.05);针刺组CORT较模型组显著降低(P〈0.05),CORT昼夜节律恢复;模型组大鼠Per2基因表达较空白组显著升高(P〈0.05),而针刺组针刺治疗后Per2基因表达显著下调,与模型组比较差异有统计学意义(P〈0.05)。结论:针刺对糖尿病抑郁模型大鼠血糖、抑郁情绪及CORT昼夜节律具有调节作用,其作用机制可能是通过下调糖尿病抑郁大鼠SCN内Per2基因表达,从而降低对正性过程的反馈抑制而实现。Objective: To observe the effects of acupuncture on circadian rhythm of CORT and m Per1/m Per2 genes expression on diabetes mellitus rats with depression so as to explore the biological mechanism of acupuncture for diabetes mellitus with depression. Methods: Thirty-six healthy SD rats were randomly divided into a blank group( 12 rats),a model group( 12 rats) and an acupuncture group( 12 rats). The rats in model group and acupuncture group were injected with STZ and treated with chronic unpredictable stress to establish the model of diabetes mellitus with depression. Rats in the acupuncture group were treated with acupuncture at "Zusanli"( ST36),"Sanyinjiao"( SP6) and "Baihui"( GV20),30 min for each time,once a day for totally 7 days. Rats in the blank group and model group were treated with identical immobilization but no treatment was given. After acupuncture treatment,blood glucose,open-field experiment and circadian rhythm of CORT and m Per1,m Per2 genes expressions were detected. Results: Blood glucose in model group was significantly higher compared with those in the blank group( P〈0. 05),while that in the acupuncture group was significantly lower than that in the model group( P〈0. 05). The numbers of horizontal movement and vertical movement in the model group were significantly lower than that in the control group( P〈0. 05),while that in the acupuncture group was significantly improved compared with that in the model group( P〈0. 05). The circadian rhythm of CORT disappeared in the model group,which was improved into normal rhythm after acupuncture treatment. The genetic expression of m Per2 in SCN in the model group increased significantly than that in the blank group( P〈0. 05),while after 7 days of acupuncture treatment,the expression level of m Per2 decreased significantly than that in the model group( P〈0. 05).
