摘  要:目的:探讨附子含药血清对大鼠原代肝细胞能量代谢的影响。方法:制备含药血清,大鼠原代肝细胞随机分为空白血清组和含药血清组。干预24 h后,分光光度计检测细胞内Na~+-K~+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性,高效液相色谱法(HPLC)检测细胞内ATP、ADP、AMP含量及细胞能荷,激光共聚焦显微镜检测线粒体膜电位。结果:与空白血清相比,附子含药血清可显著升高肝细胞Na~+-K~+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性,增加细胞内ATP含量,升高线粒体膜电位。结论:热性中药附子促进细胞能量代谢,增加细胞内能量储备。Objective: To observe the effect on energy metabolism of rat primary hepatocytes with serum containing aconite.Methods: Medicated serum was prepared on SD rat. Primary rats hepatocytes were divided into blank serum group and medicated serum group. After intervened for 24 hours,the activities of Na~+-K~+-ATPase and Ca^(2+)-Mg^(2+)-ATPase were detected with spectrophotometer. The contents of ATP,ADP,AMP were tested with high performance liquid chromatography( HPLC). Mitochondrial membrane potential was detected with laser confocal microscope. Results: Compared with blank serum group,serum containing aconite can significantly increase the activities of Na~+-K~+-ATPase,Ca^(2+)-Mg^(2+)-ATPase and the content of ATP. Mitochondrial membrane potential was also significantly improved. Conclusion: The aconite with heat nature can increase energy reserve and promote energy metabolism.
