清心滋肾汤对绝经期大鼠下丘脑β-EP和5-HT的影响

摘　　要：目的：研究中药清心滋肾汤对绝经期大鼠下丘脑中β-内啡肽（β-EP）和5-羟色胺（5-HT）含量的影响,探讨其治疗围绝经期综合征的作用机制。方法：选用自然衰老的绝经期雌性大鼠（12月龄）40只,随机分为4组,绝经期组（模型组）、清新滋肾汤高、中、低剂量组,另选4月龄雌性大鼠10只作为青年对照组。连续灌胃4周后采用酶联免疫法检测各组大鼠下丘脑中的β-EP和5-HT含量。结果：与青年组相比,模型组大鼠β-EP含量明显降低、5-HT水平明显升高,清心滋肾汤治疗后各剂量组β-EP含量均高于模型组,差异有统计学意义,而5-HT水平下降,高、中剂量组与模型组比较有统计学意义。结论：清心滋肾汤能上调绝经期大鼠下丘脑中β-EP的含量,同时下调5-HT水平,为清心滋肾汤治疗绝经期综合征提供科学依据。Objective： To explore the effects of Qingxin Zishen Decoction（QXZSD） on the levels of β-EP and 5-HT in the hypothalamus of climacteric rats and illuminate the mechanism of QXZSD treating menopausal syndrome. Methods： At the age of12 months,the climacteric rats were randomly divided into 4 experimental groups： the menopausal transition group and the QXZSD low-dose,mid-dose,high-dose groups. Ten 4-month old female rats were as young control group. The rats were given different dosages by intragastric administrations continuously for 4 weeks. Then we measured β-EP and 5-HT in the hypothalamus by ELISA. Results： The level of β-EP of the menopausal transition group was significantly lower than that of the young control group and the 5-HT was higher. The level of β-EP of the each dose group of QXZSD was higher than that of the menopausal transition group. The level of 5-HT of the each dose group of QXZSD was lower than that of the menopausal transition group. Conclusion： QXZSD can improve the level of β-EP and reduce the level of 5-HT in the hypothalamus of climacteric rats,providing a scientific basis for QXZSD treating the menopausal syndrome.
