川续断总皂苷对大鼠成骨细胞增殖、分化及OPG/RANKL mRNA表达的影响

摘　　要：目的研究川续断总皂苷对大鼠成骨细胞增殖、分化及护骨素/核因子κB受体活化因子配体(OPG/RANKL)mRNA表达的影响。方法不同浓度的川续断总皂苷作用于体外培养的大鼠成骨细胞不同时间,采用MTT法检测成骨细胞增殖功能,PNPP法检测成骨细胞中ALP活性,半定量RT-PCR法检测成骨细胞中OPG/RANKL mRNA。结果 20、50μg/mL川续断总皂苷可显著促进成骨细胞的增殖(P<0.05或P<0.01);20、50μg/mL川续断总皂苷可显著促进成骨细胞的分化(P<0.05或P<0.01);10、20、50μg/mL川续断总皂苷可显著上调成骨细胞中OPG/RANKL mRNA的比值,抑制破骨细胞的分化(P<0.01)。结论川续断总皂苷对体外培养的大鼠成骨细胞的增殖、分化及破骨细胞的分化有显著影响。Objective To study the effects of total saponins of Dipsacus asper on the proliferation and differentiation of osteoblasts and the expression of mRNA of osteoprotegerin and NF-kB receptor ligand （OPG/RANKL） in rat osteoblasts. Methods The total saponins of Dipsacus asper of different concentrations were applied to rat osteoblasts in vitro for different times. The effect on the proliferation of osteoblasts was detected by MTT assay. The effect of the medicine was detected by PNPP assay. The osteoblast OPG/RANKL mRNA was detected by the semi-quantitative RT-PCR. Results 20,50lag/mL total saponins of Dipsacus asper could significantly promote the differentiation of osteoblasts （P〈0.05 or P〈0.01）, and they could also significantly promote the proliferation of osteoblasts （P〈0.05 or P〈0.01）. The ratio of OPG/RANKL mRNA in osteoblasts was significantly increased by 10, 20, 50μg/mL total saponins of Dipsacus asper, and the differentiation ofosteoclasts was inhibited （P〈0.01）. Conclusion The total saponins ofDipsacus asper can significantly affect the proliferation and differentiation of rat osteoblasts and the differentiation of rat osteoclasts in vitro.
