摘  要:目的观察丹皮酚对蛋白聚糖诱导的小鼠关节炎模型骶髂关节滑膜细胞BMP-2、Cbfα1及Smad1mRNA表达的影响,初步从分子水平探讨丹皮酚治疗强直性脊柱炎的机理。方法将40只BALB/c小鼠随机分为正常组、模型组、柳氮磺吡啶组和丹皮酚组,每组10只。除正常组外,其余三组采用完全弗氏佐剂+蛋白聚糖腹腔注射建立蛋白聚糖诱导的BALB/c小鼠关节炎模型。实时荧光定量PCR法检测丹皮酚对滑膜细胞BMP-2、Cbfα1及Smad1mRNA表达变化的影响。结果与正常组比较,模型组的BMP-2、Cbfα1和Smad1基因表达明显升高(P<0.01);与模型组比较,柳氮磺吡啶组的BMP-2、Cbfα1和Smad1基因表达差异无统计学意义(P>0.05),而丹皮酚组的基因mRNA表达具有显著差异(P<0.05)。结论丹皮酚能显著抑制强直性脊柱炎病理性骨化,且这种抑制作用与下调滑膜细胞BMP-2、Cbfα1和Smad1基因表达,从而阻断BMP/Smad成骨信号通路有关。Objective To observe the effects of paeonol on the expression of BMP-2, Cbfα1 and Smadl mRNA in synoviocytes of arthritis model in mice induced by proteoglycan, and to explore the mechanism of paeonol in the treatment of ankylosing spondylitis from the molecular level. Methods Forty BALB/c mice were randomly divided into normal group, model group, sulfasalazine group and paeonol group. In addition to normal group, the remaining 3 groups were injected with complete Freund's adjuvant and proteoglycan to establish the BALB/c induced proteoglycan arthritis model. The effects of paeonol on the expression of BMP-2, Cbfα1 and Smadl mRNA in synovial cells were detected by real-time fluorescence quantitative PCR. Results Compared with normal group, the expression of BMP-2, Cbfα1 and Smadl in model group was significantly higher (P〈0.01). Compared with model group, there were no significant differences in the expression of BMP-2, Cbfα1 and Smadl in sulfasalazine group (P〉0.05), the mRNA expression ofpaeonol group was significantly different (P〈0.05). Conclusion Paeonol can significantly inhibit the pathological ossification of ankylosing spondylitis, and this inhibition is related to down-regulating the expression of BMP-2, Cbfα1 and Smadl genes in synovial cells, thus blocking the BMP/Smad osteogenesis signal pathway.
