摘  要:目的:建立起高效三倍体薄皮甜瓜再生体系和遗传转化体系。方法:以哈研3号薄皮甜瓜为试材,采用组织培养技术,在二倍体甜瓜再生体系的基础上,摸索外植体类型、外源调节剂种类和配比,以及各阶段的培养条件。结果:三倍体薄皮甜瓜再生最佳外植体为带有叶芽的茎段,芽诱导率为94%;芽诱导培养基最佳组合为:MS+6-BA 1.0 mg/L+KT 0.5 mg/L,Agr 1.0%;芽伸长培养基最佳组合为:MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,Agr 1.2%;壮苗培养基为:MS+6-BA 0.2 mg/L+GA30.1 mg/L,Agr 1.2%;生根培养基最佳组合为:1/2MS+NAA 0.5 mg/L,Agr 1.0%;培养条件为:25℃、14h光照/12h黑暗;确定卡那霉素的筛选浓度为2.5mg/L;确定头孢唑啉钠的除菌浓度为500 mg/L。结论:建立三倍体薄皮甜瓜高效再生体系和遗传转化体系,为今后的大量扩繁和转基因研究打下基础。Objective: To establish a high efficient triploid thin-skinned melon regeneration system and a genetic transformation system. Method: Based on the regeneration system of the diploid melon,the explant type,the type and proportion of the exogenous regulator and the culture of each stage were studied by using the tissue culture technique. condition. Result: The best explants of shoot regeneration were shoots with shoots of 94%. The optimum combination of shoot induction medium was MS + 6-BA 1. 0 mg/L +KT 0. 5 mg L,Agr 1. 0%. The optimum combination of bud elongation medium was: MS + 6-BA 0. 5 mg/L + NAA 0. 1 mg/L,Agr 1. 2%; strong seedling culture medium: MS + 6-BA 0. 2 mg/L + GA30. 1 mg/L,Agr 1. 2%; The best combination of rooting medium: 1/2 MS + NAA 0. 5 mg/L,Agr 1. 0%; culture conditions: 25 ℃,14 h light/12 h dark; determine kanamycin screening concentration of 2. 5 mg/L; The concentration of cefazolin sodium was 500 mg/L. Conclusion: The high efficient regeneration system and genetic transformation system of triploid thin-skinned melon were established,which laid the foundation for the future research of mass propagation and transgenic.
