摘  要:目的:研究虎掌南星多糖的含量及其单糖种类及摩尔比。方法:首先采用水提醇沉的方法提取虎掌南星多糖并用苯酚-硫酸法来测定其粗多糖含量;再用柱前衍生化-高效液相色谱法来测定单糖组成并通过标准回归方程计算单糖摩尔比。结果:虎掌南星多糖提取率为4.65%,含有量为56.3%,由甘露糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖组成,其单糖摩尔比为3.54∶3.36∶313.39∶9.01∶1.69。结论:苯酚-硫酸法具有显色稳定,操作简便,重复性良好等优点,可用于虎掌南星多糖含有量的测定;柱前衍生化高效液相色谱法具有简单、准确、快速、重现性好等优点,可用于虎掌南星多糖中单糖组成分析。Objective: To study the polysaccharides content of Pinellia pedatisecta, and the monosaccharide composition and mole ratio. Method: Water extraction and alcohol precipitation method was used to extract polysaccharides from Pinellia pedatisecta, and the content of polysaccharides was detected by phenol sulfuric acid method. Then Pre column Derivatization - High performance liquid chromatography was used to investigate the monosaccharide composition and the linear regression equation was conducted to calculate the molar ratio. Result: The extraction rate of Pinellia pedatisecta polysaccharides was 4.65%, and the content of polysaccharides was 56.3%. The monosaccharides of the polysaccharides were composed of mannose, galacturonic acid, glucose, galactose and arabia sugar, and the monosaccharide ratio were 3.54:3.36:313.39:9.01:1.69. Conclusion: Phenol sulfuric acid method is stabile, simple and repeatable, and it can be used for the determination of polysaccharides content of Pinellia pedatisecta. Pre column Derivatization High Performance Liquid Chromatography is simple, accurate, rapid and reproducible, and it can be used for the analysis of the monosaccharide composition of Pinellia pedatisecta polysaccharides.
