肾炎防衰液通过SPHK1/S1P信号通路对糖尿病肾病大鼠肾脏炎症的干预作用

摘　　要：目的:探讨肾炎防衰液通过鞘氨醇激酶1-1磷酸鞘氨醇(SPHK1/S1P)信号通路对糖尿病肾病大鼠肾脏炎症的干预作用。方法:采用单侧肾切除+高脂饲养喂养+链脲佐菌素(STZ)"三联"造模方法构建糖尿病肾病大鼠模型,模型组分别予肾炎防衰液和厄贝沙坦治疗8周,测定24 h尿蛋白定量及血生化指标,检测肾脏Sph K1、S1p2、TNF-α、NF-κBp65表达水平的变化。结果:肾炎防衰液可以降低DKD大鼠24 h尿蛋白定量(P<0.01),减少DKD肾组织Sph K1、S1p2、TNF-α、NF-κBp65mRNA表达(P<0.05),降低Sph K1、p NF-κBp65的蛋白表达。结论:肾炎防衰液可通过抑制Sph K1/S1P信号通路的表达,减轻肾脏炎症反应,对糖尿病肾病大鼠肾脏起到一定的保护作用。Objective：To Study the effect of Shenyanfangshuai Formula to anti - inflammation in diabetic nephropathy rats by SPHK1/S1P signaling pathway. Methods ：The model of diabetic nephropathy rat were constructed by unilateral nephrectomy, high - fat diet feeding and streptozotocin （STZ）. The model groups were given Shenyanfangshuai formula and irbesartan for 8 weeks, then determine 24 hour urinary protein, blood biochemical, SphK1, S1p2, TNF -α, NF - κB in kidney. Results： Shenyanfangshuai Formula can reduce 24 hours urine protein （ P 〈 0.01 ） and the expression of SphK1, S1 p2, TNF -α , NF - κBp65mRNA （ P 〈 0.05 ） , decrease the expression of SphK1, pNF - κBp65 protein in DKD rats. Conclusion： Shenyanfangshuai Formula may play a protective role in diabetic nephropathy rats by inhibiting the expression of SphK1/S1P signaling pathway and reducing renal inflammation.
