摘 要:为了研究迟缓爱德华氏菌刺激下牙鲆基因表达作出的响应,采用高通量测序平台(Illumina HiSeq)分别对迟缓爱德华氏菌刺激的牙鲆和未受刺激牙鲆的肾脏进行转录组测序,并对差异基因进行生物信息学分析,包括GO(基因功能注释)、SNP(单核苷酸多态性)分析、SSR(简单重复序列)分析等.结果表明,用Trinity软件对clean reads进行拼接后,聚类得到222 980条转录本,总长200 234 420 bp,平均长度898 bp,N50为1 886 bp.对组装的序列进行基因功能注释后,有99 808条序列得到了注释.筛选出2 502个差异表达比较显著的基因,表达量上调的有1 280个,下调的有1 222个.GOseq结果显示差异基因显著富集在38个GO terms上,包括6个细胞组分、12个生物学过程和20个分子功能.差异基因共获得277个KEGG通路富集,627个差异基因显著富集到20条信号通路上,其中,8个与信号通路有关,3个与发育有关,9个与相关疾病有关.To explore the response of gene expression of Paralichthys olivaceus to the Edwardsiella tarda stimulation,the transcriptome sequencing was conducted on the kidneys of stimuluted and unstimuleted P. olivaceus by E. tarda by Illumina HiSeq,and the differential genes were analyzed by bioinformatics,including GO(gene ontology),SNP(single nucleotide polymorphism)analysis,SSR(simple sequence repeats). The results showed that 222 980 transcripts were spliced by Trinity software with a total length of 200 234 420 bp,an average length of 898 bp and an N50 of 1 886 bp. After annotating the assembled genes for gene function,99 808 sequences were annotated. A total of 2 502 differentially expressed genes were screened,with 1 280 genes up-regulated and 1 222 down-regulated. The GOseq results showed that the differentially expressed genes were significantly enriched over 38 GO terms,including 6 cellular components,12 biological processes and 20 molecular functions. A total of 277 KEGG pathways were enriched and 627 differential genes were significantly enriched to 20 signal pathways,among which eight were related to the signaling pathway,three were related to development and nine were related to some diseases.
