摘  要:目的为了提高纯镁的耐腐蚀性和细胞的生物相容性,对纯镁超声微弧氧化处理后,以植酸为偶联剂载丹参素钠。方法选择小鼠成骨细胞MC3T3-E1,实验A组纯镁超声微弧氧化,B组纯镁超声微弧氧化-植酸,C组纯镁微超声弧氧化-植酸-10μg/ml丹参素钠,通过SEM扫描电镜观察试件表面形貌、检测表面元素,CCK-8检测试件表面细胞增殖和黏附能力,通过激光共聚焦显微镜检测试件表面细胞表达的骨架伸展形态。结果 3组材料表面细胞的黏附、增殖能力顺序为C组﹥B组﹥A组,各组试件表面细胞的生长和伸展情况C组最佳,B组次之,A组最低,纯镁超声微弧氧化-植酸-丹参素钠复合涂层对成骨细胞MC3T3-E1黏附、增殖有不同程度的促进作用,并提高了细胞生物活性,对A、B、C三组实验结果进行统计学分析均具有统计学意义。结论纯镁超声微弧氧化涂层通过植酸偶联载10μg/ml丹参素钠涂层的细胞相容性最佳。Objective In order to improve the corrosion resistance and cellular biocompatibility of pure Magnesium,we construct a new composite material using ultrasonic micro-arc oxidized magnesium to carry salvianic acid A sodium coupled by phytic acid. Methods The materials are grouped as followed,A,ultrasonic micro-arc oxidized pure magnesium; B,ultrasonic micro-arc oxidized puremagnesium-phytic acid; C,ultrasonic micro-arc oxidized pure magnesium-phytic acid-salvianic acid A sodium. First,the morphology and surface element of samples are observed using scanning electron microscope(SEM). Next,we test the cellular biocompatibility using mouse osteoblast cell line MC3 T3-E1. Cell adhesion and proliferation are detected on the material in each group using cell counting Kit-8(CCK-8). Finally,we use confocal microscope to characterize cytoskeleton arrangement in each group. Results Group C presents the most cellular biocompatibility,including optimal cell proliferation,adhesion and cytoskeletal extension. Group A presents the least. While group B is between the two groups. All the comparisons have significant statistic differences. In this way,ultrasonic micro-arc oxidized pure magnesium-phytic acid-salvianic acid A sodium composite layer can prominently improve the cellular biocompatibility by enhancing cell adhesion and proliferation. Conclusions The composite material layer with ultrasonic micro-arc oxidized pure magnesium-phytic acid-salvianic acid A sodium presents has the best cellular biocompatibility.
