摘 要:目的观察灯盏乙素(Scutellarin,Scu)对神经母细胞瘤(SH-SY5Y)细胞中β淀粉样蛋白(β-Amyloid peptide,Aβ)生成的影响,探讨其针对阿尔茨海默病(Alzheimer’s Disease,AD)发挥的防治作用。方法将SH-SY5Y细胞分为对照组、Aβ处理组和Aβ+Scu处理组,用CCK-8试剂盒测定细胞存活率;分别用免疫组织化学法、蛋白质印迹法和实时荧光定量聚合酶链反应法检测各组细胞Aβ_(1-42)、β-淀粉样前体蛋白(β-amyloid precursor protein,APP)、APP切割酶1(β-Site APP-leavingnzymes1,BACE1)、APP切割酶2(β-Site APP-leavingnzymes2,BACE2)的表达。结果与对照组相比,Aβ处理组的细胞存活率下降,Aβ_(1-42)、APP、BACE1蛋白和m RNA的表达水平增高,BACE2蛋白和m RNA的表达水平下降,Scu干预后,各检测指标的变化与Aβ处理组的结果正好相反。结论 Scu能够影响Aβ生成相关因子的表达,可能通过减少Aβ的沉积在AD防治中发挥积极作用。Objective The aim of this study was to investigate the effect of scutellarin(Scu) on generation of β-amyloid protein(Aβ) in SH-SY5 Y cells,and to reveal the positive prevention and treatment effect of the drug to Alzheimer 's disease(AD).Methods SH-SY5 Y cells were divided into control group,Aβ group and Aβ +Scu group.Cell counting kit-8(cck8) was employed to detect cell survival rate.The expression of Aβ_(1-42),β-amyloid precursor protein(APP),β-site APP-leavingnzymes1(BACE1),β-site APP-leavingnzymes2(BACE2) were examined by immunohistochemistry,western blotting and real-time PCR,respectively.Results Compared with the control group,the cells survival rate was decreased,the protein and m RNA expression of Aβ_(1-42),APP and BACE1 were increased,and the protein and m RNA expression of BACE2 were decreased in the model group.After treatment of Scu,the changes of each test index were just the opposite compared with the Aβ group.Conclusion Scu might affect the expression of correlated factors of Aβ generation and play a positive role in the prevention and treatment of AD ppossibly by reducing the deposit of Aβ.
