摘 要:研究旨在探讨黄芩苷对热应激小鼠卵巢抗氧化和细胞凋亡的作用机制。将小鼠分为常温对照组(生理盐水)(C组)、黄芩苷(50 mg/kg体重)组(C+B组)、41℃热应激组(H组)和热应激加黄芩苷(50 mg/kg体重)组(H+B组),连续注射7 d,第8天41℃热应激2 h,立即处死,取卵巢组织进行切片观察并检测丙二醛(MDA)和一氧化氮(NO)的含量及总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)的活力。Western Blot检测Nrf2、Keap1、HO-1和NQO1蛋白表达。免疫组化法检测Bcl-2、Bax、Caspase 3蛋白表达。结果显示,热应激引起小鼠卵巢组织氧化损伤。黄芩苷能显著降低组MDA和NO含量,显著或极显著增加SOD、CAT和GSH-Px活力。此外,热应激可显著增加Nrf2、Keap1、HO-1和NQO1蛋白表达,黄芩苷干预后Nrf2等蛋白的表达降低。黄芩苷干预后Bax蛋白表达显著降低,Bcl-2、Caspase 3蛋白表达显著升高。上述表明,黄芩苷能缓解热应激下小鼠卵巢组织的氧化损伤及细胞凋亡,其机制可能与提高抗氧化能力及Nrf2蛋白的表达有关。To study the effect of baicalin on oxidative damage of ovary in heat stressed mice,and to explore its possible antioxidant mechanism,we randomly divided the The mice into four groups including normal control group( C),baicalin group( C + B),heat stress group( H) and baicalin pretreatment and heat stress group( H + B). Mice were intraperitoneally administered with baicalin or sham solution for 7 days,and then were heat-stressed at 41℃ on day 8. MDA and NO contents,T-SOD,GSH-PX and CAT activities were analyzed using their corresponding kits. Nrf2,Keap1,HO-1 and NQO1 protein expressions were detected using western-blot technique. The expression of Bcl-2,Bax,and Caspase 3 protein was detected by immunohistochemistry. Oxidative damage was observed in ovarian tissue of the mice after thermal stress. The contents of MDA and NO in the heat stress with baicalin group were significantly decreased while the activities of SOD,CAT and GSH-Px were significantly increased. In addition,baicalin significantly decreased Nrf2,Keap1,HO-1 and NQO1 protein expressions. Baicalin also significantly decreased the expression of Bax protein and significantly increased the expressions of Bcl-2 and Caspase 3 protein. It showed that baicalin protected mice ovarian from heat-stress induced oxidative damage and apoptosis possibly by increasing the expressions of antioxidant enzymes and Nrf2 protein.
