CTGF对犬眼小梁切除术后滤过泡瘢痕化和眼压的影响

摘　　要：为了检测犬眼小梁切除术后滤过泡区域组织中结缔组织生长因子(CTGF)的表达量,并结合外源性注入CTGF探索CTGF对犬眼小梁切除术后滤过泡瘢痕形成和眼压的作用。方法:(1)选取体质大致相同的本地杂交犬12只。左眼行小梁切除术为试验组,右眼为对照组。术后不同时间取滤过泡区域组织,进行免疫组织化学染色并分析;(2)选取体质大致相同的本地杂交犬16只左眼行小梁切除术,随机分为试验组和对照组,结膜瓣下分别注射等量的CTGF和生理盐水,术后每天观察滤过泡生成情况并测量术前以及术后术眼眼压。结果:(1)试验组中CTGF蛋白阳性颗粒的阳性面积率和光密度(OD)值随手术后时间变化呈先升高后下降的变化,在第7天达到峰值。对照组中CTGF蛋白阳性颗粒和OD值几乎为0;(2)试验组平均滤过泡失败时间极显著低于对照组(P<0.01)。试验组比对照组眼压恢复快,差异极显著(P<0.01)。表明CTGF表达水平在小梁手术后呈先升高后下降的变化,7 d达峰值。当外源性注入CTGF,加速了滤过泡瘢痕形成和眼压升高,导致手术失败。The amount of connective tissue growth factor（ CTGF） expressed in filter bleb organizations was detectived during filter bleb scarring process after canine ocular trabeculectomy and then researching the effection of CTGF for the filter bleb scarring after canine ocular trabeculectomy by injection of exogenous CTGF to study the role of CTGF in canine glaucoma surgery.（ 1）. 12 local hybrid dogs were selected to perform the ocular trabeculectomy at their left eyes as the experimental group,and the right eyes were maintained as a control group. the bleb regional organization of two eyes of any two dogs were clipped to made biopsy for immunohistochemistry straining,then electron microscopy was used to image and quantitative analysis.（ 2）. 12 same constitution local hybrid dogs were selected to perform the ocular trabeculectomy at their left eyes,then it were randomly allocated to the experimental group and the control group. Thereafter,the same amount of CTGF and saline were injected under conjunctival flap,then filter blebs were observed daily to record the survival situation and the failure time. Preoperative and postoperative IOP were measured and recorded.（ 1）. In test group,Protein positive area ratio of CTGF Positive surface protein particles and optical density（ OD） value of CTGF protein particles presented the change that increased then decreased as the time after surgery change. On the 3 rd day it accelerated obviously,the 7 th day it accelerated mostly. In blank control group,Protein positive area ratio of CTGF Positive surface protein particlesthe and OD value of CTGF protein particles were very small.（ 2） Compared with control group,the average filtering bleb failed time of test group was decteased significantly（ P〈0. 01）. Compared with control group,intraocular pressure of test group quick recovery,and the difference was significantly（ P〈0. 01）.
