摘 要:目的探索以维生素E琥珀酸酯为乳化剂制备阿霉素纳米乳的方法和条件。方法分别筛选优化阿霉素和维生素E琥珀酸酯比例、搅拌速度及搅拌时间、纳米乳中药物浓度,寻找制备阿霉素-维生素E琥珀酸酯纳米乳的最优条件,并采用紫外分光光度法分析阿霉素与维生素E琥珀酸酯的相互作用。结果通过筛选优化,确定了制备的最佳条件:阿霉素和维生素E琥珀酸酯为1∶2,阿霉素浓度为5%,搅拌速度400r·min^(-1),搅拌时间为12h。结论利用维生素E琥珀酸酯的乳化和对阿霉素减毒增效两方面作用,初步制备了阿霉素-维生素E琥珀酸酯纳米乳。制备方法简单,试剂耗费少,有望成为雹霉素药物的一种制剂途径。OBJECTIVE To explore the methods and conditions of preparation of adriamycin nanoemulsion with vitamin E succinate as emulsifier. METHODS The selection of adriamycin and vitamin E succinate ratio, stirring speed and stirring time, drug concentration in nanoemulsion to find the optimal conditions for the preparation of adria- mycin-vitamin E succinate nanoemulsion, and UV analysis of interaction of adriamyein and vitamin E succinate by spectrophotometry. RESULTS The optimum conditions for the preparation were determined by the optimization of screening method : adriamycin and vitamin E succinate was 1 : 2, the concentration of adriamycin was 5 %, the stirring speed was 400r · min-1, and the stirring time was 12h. CONCLUSION Vitamin E succinate nanoemulsion was prepared by emulsification of vitamin E succinate and its synergistic effect on adriamyein in two aspects. The preparation method is simple and the reagent consumption is low, which is expected to be a preparation method of adriamycin drugs.
