益气化瘀方对子宫复旧不全大鼠子宫内膜细胞凋亡与产后子宫内膜增殖影响研究

摘　　要：目的:通过对比分析益气化瘀方对子宫复旧不全SD级大鼠子宫内膜细胞凋亡及子宫内膜增殖影响的研究,为孕妇产后疾病的治理提供理论依据。方法:选取试验用无特定病原体级实验动物(SPF级动物)SD大鼠为研究对象,按照实验需求,分为5组,除去空白组以外的四组进行子宫内接种大肠杆菌,进行造模,然后给药,观察大鼠子宫内膜细胞凋亡率、大鼠子宫内膜细胞凋亡因子Bcl-2和Bax的表达情况。结果:(1)与模型组对比,新生化颗粒组、益气化瘀方低剂量组和益气化瘀方高剂量组大鼠子宫内膜凋亡率降低,P<0.05;与新生化颗粒组比较,益气化瘀方高剂量组大鼠子宫内膜细胞凋亡率无统计学差异,P>0.05,益气化瘀方低剂量组大鼠子宫内膜凋亡率升高,P<0.05;与空白组比较,模型组大鼠子宫内膜凋亡率显著升高,P<0.05。(2)与模型组对比,新生化颗粒组、益气化瘀方低剂量组和益气化瘀方高剂量组Bcl-2蛋白表达增加,Bax表达减少,P<0.05;与新生化颗粒组比较,益气化瘀方高剂量组Bcl-2蛋白表达和Bax表达无统计学差异,P>0.05,益气化瘀方低剂量组Bcl-2蛋白表达减少,Bax表达增加,P<0.05;与空白组比较,模型组Bcl-2蛋白表达明显下降,Bax表达增加,P<0.05。结论:益气化瘀方能通过调节使Bax蛋白表降低,使Bcl-2蛋白表达增加,从而降低子宫内膜细胞凋亡率,促进产后子宫增殖恢复。Objective： To evaluate the effect of Yiqi Huayu Decoction on the apoptosis of endometrial cells and the proliferation of endometrium in SD rats with gradeⅠ,and to provide theoretical basis for the treatment of postpartum diseases. Methods：Sprague-Dawley（ SD） rats were gathered as experimental subject. The rats were randomly divided into five groups according to the experimental requirements. And then made model among the four groups except the blank group. The expression of Bcl-2 and Bax in rat endometrial cells was observed. Rsults：（ 1） Compared with the model group,the apoptotic rate of the endometrium was decreased in the low dose of Yiqi Huayu Decoction group,the high dose of Yiqi Huayu Decoction group,P0. 05,and the New Biochemical Particles group. There was no significant difference in the rate of endometrial cell apoptosis in the high dose of Yiqi Huayu Decoction group（ P 0. 05）. The apoptotic rate of endometrium in rats with low dose of Yiqi Huayu Decoction increased,P0. 05. Compared with the blank group,the apoptosis rate of the endometrium in the model group was significantly higher than that in the blank group（ P0. 05）.（ 2） Compared with the model group,the expression of Bcl-2 protein increased and the expression of Bax were decreased in the New Biochemical Particles group,the low dose Yiqi Huayu Decoction group and the high dose of Yiqi Huayu Decoction group,P0. 05. Compared with the New Biochemical Particles group,the expression of Bcl-2 and Bax had no significant different in the high dose of Yiqi Huayu Decoction group. The expression of Bcl-2 deceased and Bax increased in the low dose of Yiqi Huayu Decoction group（ P0. 05）. Compared with the blank group,the expression of Bcl-2 protein in the model group decreased significantly and the expression of Bax increased,P0. 05. Conclusion： Yiqi Huayu Decoction can reduce the expression of Bcl-2 protein by regulating the Bax protein expression,thus reduce the rate of endometrial cell apoptosis and promote the recovery of po
