活血宁心解毒方对急性心肌缺血模型大鼠心电图、血流动力学及氧化应激指标的影响  

Influence of Huoxue Ningxin Jiedu Decoction on Electrocardiogram,Hemodynamic and Oxidative Stress Index in Acute Myocardial Ischemia Rats

摘　　要：目的观察活血宁心解毒方对急性心肌缺血模型大鼠心电图、血流动力学及氧化应激指标的影响并探讨其机制。方法 50只Wiatar大鼠随机分为5组,每组10只,分别为空白对照组、模型对照组、活血宁心解毒方低剂量组、活血宁心解毒方中剂量组和活血宁心解毒方高剂量组,除空白对照组外采用结扎冠状动脉前降支法制备急性心肌缺血模型,空白对照组和模型对照组给予蒸馏水灌胃,活血宁心解毒方低中高剂量组分别给予中药煎煮液灌胃。2周后观察各组大鼠心电图变化、血流动力学、凝血及氧化应激指标。结果与模型组比较,活血宁心解毒方组可减轻炎性细胞浸润、血管扩张、心肌纤维化水肿。与模型组比较,活血宁心解毒方组大鼠心电图ST段抬高值明显减少,且不同组间比较差异均有统计学意义(P<0.05)。活血宁心解毒方组左房平均压(mLAP)、平均二尖瓣压力差(mMPG)、肺动脉平均压(mPAP)显著降低,且活血宁心解毒方剂量越高,mLAP、mMPG和mPAP越低(P<0.05);活血宁心解毒方组大鼠全血黏度显著降低(P<0.05);活血宁心解毒方组大鼠超氧化物歧化酶和谷胱甘肽过氧化物酶升高,脂质过氧化物降低,且呈剂量依赖性(P<0.05)。结论活血宁心解毒方可逆转急性心肌缺血模型大鼠的心电图、血流动力学、凝血及氧化应激指标,改善急性心肌缺血。Objective： To study the influence of Huoxue Ningxin Jiedu Decoction on electrocardiogram,hemodynamic and oxidative stress index in acute myocardial ischemia rats. Methods： 50 Wiatar rats were randomly divided into 5 groups,10 in each： the blank control group,model control group,low dose group of Huoxue Ningxin Jiedu Decoction,middle dose group of Huoxue Ningxin Jiedu Decoction and high dose group of Huoxue Ningxin Jiedu Decoction respectively. Except the blank control group,the other groups were adopted the anterior descending coronary artery ligation of preparation to establish acute myocardial ischemia model. The blank control group and the model control group were given distilled water to fill the stomach. Huoxue Ningxin Jiedu Decoction groups were given traditional Chinese medicine decoction lavage. After 2 weeks,the electrocardiogram changes,hemodynamics,blood clotting and oxidative stress index were observed. Results： Compared with the model group,inflammatory cell infiltration,vasodilation and myocardial fibrosis edema reduced in Huoxue Ningxin Jiedu Decoction groups. Compared with the model group,electrocardiogram ST segment elevation value decreased significantly in Huoxue Ningxin Jiedu Decoction groups;the differences between different groups had statistical significance（P 0.05）. The mLAP,mMPG and mPAP in Huoxue Ningxin Jiedu Decoction groups were significantly reduced,and the higher Huoxue Ningxin Jiedu Decoction dose was,the lower mLAP,mMPG and mPAP were;the differences were statistically significant（P 〈 0.05）;the viscosity of whole blood in Huoxue Ningxin Jiedu Decoction groups was decreased significantly;the difference had statistical significance（P 〈 0.05）;the levels of SOD and GSH-PX were increased,and LPO were decreased in Huoxue Ningxin Jiedu Decoction groups,which was dose-dependent,and the differences were statistically significant（P 〈 0.05）. Conclusion： Huoxue Ningxin Jiedu Decoction can reverse electrocardiogram,hemodynamics,blood clotting and oxidative stress
