摘 要:目的观察四妙勇安汤对大鼠尾动脉血管吻合术后吻合口再狭窄及血管内皮生长因子(VEGF)蛋白的作用。方法雄性SD大鼠随机分为两组,每组12只,均采用尾动脉离断后行显微镜下吻合手术造模,造模后12h进行干预,实验组予四妙勇安汤灌服,对照组予0.9%氯化钠注射液灌服。分别于造模后第3日和第7日的末次灌胃2h后分批留取血管组织标本,HE染色观察、免疫组织化学法检测VEGF蛋白表达,并采用图像分析软件IPP 6.0(Image-Pro-Plus 6.0)进行分析。结果与对照组比较,四妙勇安汤组在术后第3日和第7日时的血管内膜面积均明显增加(P<0.05),VEGF蛋白表达升高(P<0.05),而吻合口血管狭窄程度减低(P<0.05)。结论四妙勇安汤能促进血管吻合术后血管内皮细胞增殖,并通过调节VEGF蛋白水平的变化,加快血管修复,达到预防吻合口再狭窄的作用。Objective: To studythe effect of Simiao Yongan Decoction on anastomotic restenosis and expression of VEGF protein in rats with microvascular anastomosis. Methods: Male SD rats were randomly divided into two groups(n=12). All ofthem were treated withthe anastomosis underthe microscope afterthe tail artery were cut off. Afterthe model was established,Simiao Yongan Decoction and normal saline was respectively performed. Onthe 3rd day andthe 7th day afterthe operation,the blood vessel tissue samples were collected at 2h afterthe last Feeding. Each group was divided into two groups again. Vascular tissue specimens were fixed for HE staining observation ofhistomorphological changes and immunohistochemical detection of VEGF protein expression. Image analysis software IPP 6.0(Image-Pro-Plus 6.0) was used to analysis of pictures. Results: As compared withthe control group,the vascular intimal area andthe expression level of VEGF in Simiao Yongan Decoction group were significantly increased onthethird and seventh day after operation(P 〈 0.05),whilethe PA/EELA ratio was decreased(P 〈 0.05). Conclusion: Simiao Yongan Decoction can regulatethe proliferation ofthe vascular endothelial cell andhas positive function in repairingthe microvascular anastomosis and preventing restenosis of anastomosisthroughthe method of regulatingthe expression ofthe VEGF protein.
