摘 要:目的:采用经典的小鼠胃排空和小肠推进实验,观察小半夏汤对胃肠运动的影响。方法:采用新斯的明建立小鼠胃肠运动亢进模型,阿托品、肾上腺素、多巴胺分别建立小鼠胃肠运动抑制模型,以胃残留率和小肠推进率为指标,观察不同浓度小半夏汤对小鼠胃肠运动抑制和亢进模型的影响。结果:(1)小半夏汤高、中剂量可拮抗新斯的明引起的小鼠胃肠运动亢进,抑制其胃排空和小肠推进,小半夏汤低剂量仅可抑制新斯的明所致小肠推进加快,对胃排空无影响(P〉0.05);(2)小半夏汤高、中、低三个剂量都不能拮抗阿托品引起的小肠推进抑制(P〉0.05),但小半夏汤中剂量可显著降低阿托品模型动物的胃残留率(P〈0.05);(3)小半夏汤中、低剂量可拮抗肾上腺素所致小鼠胃肠运动抑制,显著降低小鼠胃残留率,促进小鼠小肠推进(分别P〈0.01、P〈0.05);(4)小半夏汤高、中、低剂量均能显著促进多巴胺模型小鼠的小肠推进(分别P〈0.001、P〈0.01、P〈0.01),小半夏汤中、低剂量可明显降低模型小鼠的胃残留率(P〈0.01)。结论:小半夏汤可拮抗新斯的明所致的小鼠胃排空和小肠推进亢进,同时改善阿托品引起的胃排空抑制,拮抗多巴胺和肾上腺素所致的小鼠胃排空和小肠推进抑制。提示小半夏汤对胃肠运动具有双向调节作用。其作用机理可能与胆碱能系统和肾上腺素能系统有关。Objective: Gastric emptying and small intestinal propulsion effect of Xiao Banxia Decoction( XBXD) was investigated in mice. Methods: Mice with food deprived for 18 hours were orally administered a certain amount of test meal( ca. 0. 8 g) equaling 0. 8 m L in volume. The percentage of 0. 8 g test meal remaining in the stomach after 20 minutes was estimated. Results: It showed that XBXD significantly antagonized neostigmine-induced( 2 mg/kg,p. o.) gastrointestinal motility hyperfunction. It also significantly antagonized dopamine-( 0. 56 mg/kg,i. p.),atropine-( 0. 3 mg/kg,i. p.) and adrenaline( 0. 3 mg/kg,i. p.)induced gastric emptying inhibition and small intestinal propulsion inhibition. XBXD can antagonize atropine-induced( 0. 3 mg/kg,i. p.) gastric emptying inhibition,not the small intestinal propulsion inhibition. Conclusion: XBXD can antagonize neostigmine-caused gastric emptying and small intestine hyperfunction,while improving atropine-induced gastric emptying inhibition,antagonizing dopamine and adrenaline-induced gastric emptying and small intestine propulsion. Suggesting that Xiao Banxia Decoction has a two-way regulation of gastrointestinal motility. The mechanism of action may be related to the cholinergic system.
