摘 要:目的:研究芪参益气滴丸预适应对缺氧复氧损伤心肌细胞的延迟保护作用,并进一步探讨其作用机制及与热休克蛋白70(HSP70)表达的关系。方法:建立心肌细胞缺氧复氧损伤(H/R)及缺氧预适应(HPC)模型,以芪参益气滴丸预适应(QS)与HPC分别或同时作用于心肌细胞,并加入N-乙酰半胱氨酸(NAC)作为抑制剂取消HPC的延迟保护(DP)效应,在培养24小时后对心肌细胞进行H/R损伤,通过镜下观察细胞形态,检测CCK-8、乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)以及HSP70的蛋白含量和m RNA表达,观察心肌细胞的损伤程度及HSP70的变化。结果:在经过H/R损伤后,QS组与HPC组心肌细胞表现出很好的细胞活性与抗氧化力,损伤轻(P<0.05),HSP70蛋白含量和m RNA表达均高于H/R组(P<0.5);NAC抑制了HPC对心肌细胞的延迟保护作用,表现为细胞活性降低、抗氧化能力减弱,损伤较重(P<0.05),HSP70蛋白含量及m RNA表达显著降低(P<0.05);HPC+NAC+QS组心肌细胞表现出很好的活性与抗氧化能力,损伤轻(P<0.05),HSP70蛋白含量和m RNA表达显著升高(P<0.5)。结论:芪参益气滴丸预适应对缺氧复氧损伤心肌细胞具有延迟保护作用,其作用效果与缺氧预适应相当,其作用机制与多途径上调HSP70的表达有关。Objective: To discuss delayed protection of Qi Shen Yi Qi dripping pills(QSYQ)-induced preconditioning to hypoxia/reoxygenation injury of myocardial cells, and to further discuss the connections between its mechanism and heat shock protein 70(HSP70) expression. Methods: After establishing mycardial cells of hypoxia/reoxygenation injury(H/R) and hypoxia preconditioning(HPC), Qi Shen Yi Qi dripping pills preconditioning and HPC were used to act on myocardial cells respectively or simultaneously, and N-acetyl-L-cysteine(NAC) was added as the inhibitor to cancel delayed protection effects of HPC, H/R injury was given to myocardial cells after they were cultivated for 24 hours, HSP70 protein contents and m RNA expressions, SOD, MDA, LDH and CCK-8 were detected by microscopic observation of cellular morphology, the changes of HSP70 and the injury of myocardial cells were observed. Results:After experiencing H/R injury, myocardial cells in QS group and HPC group performed the best cell activity and anti-oxidation with light injury(P〈0.05), HSP70 protein contents and m RNA expressions were higher than these of H/R group(P〈0.05); NAC inhibited the delayed protection of HPC to myocardial cells, which performed low cell activity, anti-oxidation ability weakening and severe injury(P〈0.05), HSP70 protein contents and m RNA expression lowered notably(P〈0.05); myocardial cells in HPC+NAC+QS group showed better cell activity and anti-oxidation with light injury(P〈 0.05), HSP70 protein contents and m RNA expression lifted notably(P〈 0.05). Conclusion:Qi Shen Yi Qi dripping pills preconditioning shows delayed protection to myocardial cells with H/R injury, and its effects are equivalent to these of HPC, and its mechanism might be related to up regulating HSP70 expressions by many ways.
