摘　　要：目的探讨A3亚型血型的血清学和分子生物学特征。方法采用单克隆抗-A、抗-B、抗-A1血型试剂检测2例ABO正反定型不符先证者的红细胞ABO抗原,采用标准A、B、O反定型细胞检测先证者血清中的抗-A和(或)抗-B抗体,采用PCR-序列特异性引物分析法确定基因型,采用基因测序法检测ABO基因第6内含子和第7外显子基因序列,同时采集先证者1妻子、女儿的血液标本进行血清学血型和基因检测。结果先证者1、2红细胞与抗-A呈混合视野反应,血清与B型红细胞呈4+,PCR-序列特异性引物分析基因型分别为A307/O02和A102/O01,基因测序结果第7号外显子分别为C745T和C467T突变;先证者1妻子、女儿血型分别为A型、O型,基因型分别为A101/O02、O02/O02。结论 A3亚型ABO等位基因存在遗传的多态性。Objective To explore the serological and molecular biological features of A3 subtypes.Methods ABO antigen in erythrocyte of two probands with inconformity stereotypes in positive and negative were tested by monoclonal anti-A,anti-B and anti-A1 blood type reagents,the anti-A and anti-B in serum of the probands were detected by standard A,B and O antitype cells,and the genotype was determined by using polymerase chain reaction-sequence specific primer（PCR-SSP）.The introns 6 and exon 7 of ABO gene sequences were tested by gene sequencing,and the blood specimens of the wife and daughter of the proband 1 were collected and detected by serological and genetic testing.Results The reactions of RBC of proband 1 and proband 2 with anti-A were mixed field agglutination,the reactions of the serum with B antigen were ＋ ＋ ＋ ＋,the genotypes were A307/O02 and A102/O01 respectively with the test of PCR-SSP,and the sequencing of exon 7 were C745 T and C467 T mutation respectively.The blood types were type A and type O while the genotypes were A101/O02 and O02/O02 respectively in the wife and daughter of proband 1.Conclusion There was heredity polymorphism in the allele of A3 subtype in ABO.
