摘  要:目的研究变应性鼻炎(AR)模型中鼻黏膜及肺组织中气道重塑相关细胞因子的表达,进一步探讨上下呼吸道一致性的气道重塑病理过程。方法 20只雄性大鼠随机分为AR模型组和正常对照组,每组10只。AR模型建立后,通过ELISA法检测鼻黏膜、肺组织匀浆及血液标本中转化生长因子-β1(transforming growth factor-β1,TGF-β1)、白细胞介素4(interleukin-4,IL-4)、IL-13表达,显微镜下观察并计数鼻黏膜和肺组织中嗜酸性粒细胞和肥大细胞浸润程度。结果 AR模型组鼻黏膜及肺组织中嗜酸性粒细胞以及肥大细胞与对照组相比有统计学差异(P均<0.05),但AR模型组肺组织中未出现明显的气道重塑病理改变。AR模型组鼻黏膜及肺组织中TGF-β1、IL-4、IL-13与对照组相比有统计学差异(P均<0.05),但其在血液中表达水平两组相比无统计学差异(P>0.05)。鼻黏膜与肺组织中TGF-β1、IL-4、IL-13表达的相关性具有统计学意义。结论 AR模型中鼻黏膜及肺组织中气道重塑相关细胞因子TGF-β1、IL-4、IL-13表达明显增高,且鼻黏膜及肺组织中具有一致性的表达,上下呼吸道可能具有类似的细胞因子发病机制。OBJECTIVE To study the expression of cytokines related to airwayremodeling in nasal mucosa and lung tissue in allergic rhinitis(AR) models, and to investigate the pathological process of airway remodeling in the upper and lower respiratory tract. METHODS 20 male guinea pigs were randomly divided into two groups: AR model group and normal control group (10 rats in each group). Tissue homogenate and blood samples were managed by the Elisa test to detect the expression of TGF-β1, IL-4, IL-13. Eosinophils and mast cells were observed under microscope and counted in the nasal mucosa and lung tissue infiltration. RESULTS Statistical significance was found in eosinophils and mast ceils infiltration of nasal mucosa and lung tissue in model group of AR compared with the control group(P〈0.05), but the AR group did not show obvious pathological changes of airway remodeling. There was statistical significance in TGF-β1, IL-4 and IL-13 expression of nasal mucosa and lung tissue between model group of AR and control group(P〈0.05). However their blood expression difference was not significant between the two groups (P〉0.05). Expression of TGF-β1, IL-4, IL-13 of nasal mucosa were proved to relate to those of lung tissue. CONCLUSION TGF-β1, IL-4, IL-13 expression related to airway remodeling were increased in nasal mucosa and lung tissue of AR model, which were consistent in both the nasal mucosa and lung tissue. The study suggest that the common cell factors may play similar role in the pathological process of the upper and lower respiratory tract.
