摘 要:为探究诱导β干扰素包含TIR结构域的接头分子(TIR-domain-containing adaptor inducing interferon-β,TRIF)是否参与赤眼鳟(Squaliobarbus curriculus)抗病免疫反应,克隆获得了2 263 bp的赤眼鳟TRIF基因(Sc TRIF)全长c DNA.研究结果显示Sc TRIF具有TIR结构域,Sc TRIF和同属亚罗鱼亚科的草鱼TRIF聚为一支.该基因在赤眼鳟12个组织中均有表达,其中鳃中表达水平最高.感染草鱼呼肠弧病毒后,头肾和脾脏组织中Sc TRIF表达水平均呈现先下降后上调的趋势.研究表明Sc TRIF参与赤眼鳟抗GCRV病毒的免疫应答反应.In order to investigate the potential role of TIR-domain-containing adaptor induced interferon-β receptor of Squaliobarbus curriculus( Sc TRIF) that plays in anti-disease immune reaction,the full c DNA sequence of Sc TRIF( 2 263 bp) was cloned. Our research results in this paper showed that Sc TRIF protein possesseda typical structure of TIR domain,and Sc TRIF was clustered together with that of Ctenopharyngodon Idella,belonging to the subfamily of Leuciscinae. The expression of Sc TRIF was detected in 12 tissues,with the highest expression level in gill. After GCRV infection,the Sc TRIF expression level showed an overall tendency of firstly down-regulated and then increased,in both the head kidney and spleen tissues. Our results indicate that Sc TRIF involved in the immune response of Squaliobarbus curriculus against GCRV.
