摘  要:本文探讨了免疫隔离装置PES多孔细管对胰腺INS-1细胞系及胰岛在高糖环境下的响应能力。实验采用胰岛素体外分泌法(GSIS)检测分析胰腺肿瘤细胞INS-1和大鼠胰岛组织在植入与非植入孔径为100nm的PES多孔细管两种情况下对高糖环境的响应水平。结果显示植入PES多孔管中的INS-1细胞和胰岛细胞相较于未植入PES多孔管中的INS-1细胞和胰岛细胞对高糖环境有较好的响应性,提示INS-1细胞系可以作为模拟胰岛细胞功能提供便利;同时PES多孔细管装置可作为潜在的免疫隔离装置在胰岛移植方面有更长远的应用。In this paper,the response ability of the immune isolation device(PES) to the pancreatic INS-1 cell line and islet in high glucose environment was investigated.Insulin secretion in vitro(GSIS) was used to detect the response level of pancreatic tumor cells INS-1 and rat islet tissues to high glucose environment in two cases,which were implanted with PES porous tubules with a diameter of 100 nm.Results show that implantation of PES porous tube in INS-1 cells and islet cells compared with and without PES porous tube INS-1 cell and islet cell responds well to high glucose environment,suggesting that INS-1 cells can be used as a simulation of the function of islet cells and provide convenience;PES porous tube device can be used as immune isolation device have the potential application long term in islet transplantation.
