摘  要:目的:以中医理论为指导,依据射干临床应用病种,探讨射干提取物对合胞病毒(RSV)感染豚鼠气道炎症白介素-4(IL-4)、干扰素-γ(IFN-γ)、白三烯-4(LTC4)表达的影响。方法:实验分空白组、模型组、给药组,每组10只,除空白组外,其余各组豚鼠在乙醚浅麻后鼻腔滴注100TCID50的RSV悬液100μL(每天滴鼻2次,早晚各1次,空白组给予相同剂量的5%DMEM培养液),连续给药1周。收集肺泡灌洗液(BALF)计数,并测定肺泡灌洗液中IL-4、IFN-γ、LTC4水平变化,HE染色观察肺组织形态学变化。结果:射干提取物的干预治疗能显著降低小鼠BALF中炎性细胞总数;BALF上清液中IFN-γ水平明显升高,IL-4、LTC4水平显著下降,与RSV感染模型组比较有显著性差异(P<0.05)。结论:射干提取物能明显降低豚鼠模型BALF中炎性细胞数量,影响细胞因子水平变化,减轻肺组织炎症。This study was aimed to investigate the effect of Belamcanda Chinensis extract on the expression of interleukin-4(IL-4),interferon-γ(IFN-γ)and leukotriene-4(LTC4)in airway infection induced by respiratory syncytial virus(RSV)in guinea pig based on traditional Chinese medicine(TCM)theories.There were the blank group,model group and the drug administration group,with 10 guinea pigs in each group.Except for the blank group,the remaining guinea pigs were infused with 100 μl of RSV suspension of 100 TCID50(twice daily for 1 day,once in the morning and once in the evening,and the blank group was given the same dose of 5% DMEM medium).The drug administration was continuous given for 1 week.Bronchoalveolar lavage liquid fluid(BALF)was collected and counted.Levels of IL-4,IFN-γ and LTC4 were measured.Morphological changes of lung tissues were observed by HE staining.The results showed that the intervention of Belamcanda Chinensis extract can significantly reduce the total number of inflammatory cells in BALF.The level of IFN-γ in BALF supernatant was significantly increased and the levels of IL-4 and LTC4 were significantly decreased compared with the RSV infection model group(P〈 0.05).It was concluded that Belamcanda Chinensis extract can significantly reduce the number of inflammatory cells in BALF in guinea pig model,affect changes of cytokine levels and reduce the inflammation of lung tissues.
