摘  要:以黄樟秋稍带腋芽茎段为外植体,建立并优化了黄樟组培繁育体系,研究基本培养基、不同激素及配比对黄樟茎段不定芽诱导、组培苗增殖及生根的影响。结果表明:黄樟带腋芽茎段最佳启动培养基配方为改良MS+4.0 mg/L 6-BA+0.5 mg/L NAA,在此培养基中不定芽培养7 d开始萌动,诱导率可达76.67%;不定芽继代培养基的最适宜配方为改良MS+2.0 mg/L 6-BA+0.5 mg/L NAA,在此培养基中不定芽生长健壮,增殖系数为3.78;幼苗生根培养基的最适宜配方为改良1/2 MS+0.5 mg/L IBA+0.5 mg/L IAA,在此培养基中幼苗培养13 d开始发根,生根率达85.56%。上述组织培养技术条件,可有效提高黄樟茎段的不定芽诱导率和生根率,为黄樟优良品系的工业化育苗奠定了基础。To establish a tissue culture system for Cinnamomum porrectum(Roxb)Kosterm,the stems with buds of 2-year-old C.porrectum tree were used as explants in vitro rapid propagation.Effects of different basal medium and hormone on buds initiation,shoot proliferation and rooting of tissue cultured seedlings were analyzed.The results showed that the optimal medium for inducing axillary buds was modified MS + 2.0 mg/L 6-BA + 0.5 mg/L NAA.In this medium,the start time of adventitious bud induction was 7 d and the budding rate was 76.67%.The best medium for propagation of adventitious buds was modified MS + 2.0 mg/L 6-BA + 0.5 mg/L NAA,in which the adventitious buds grew vigorously with a proliferation rate of 3.78.The best medium for rooting was modified 1/2 MS + 0.5 mg/L IBA + 0.5 mg/L IAA,in which rooting rate of the aseptic seedling reached 85.56%.This optimized tissue culture condition effectively improves the introduction rate of adventitious buds and the rooting rate of aseptic seedlings,providing a technical foundation for industrialized seedlings of improved varieties of C.porrectum.
