摘 要:目的建立丹参药材的HPLC特征图谱,并对其活性成分进行含量测定。方法采用Capcell C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.05%磷酸水作为流动相进行梯度洗脱,流速0.8 ml/min,检测波长270 nm,柱温30℃;分别对10批丹参药材的活性成分进行含量测定。结果初步建立了丹参药材的HPLC特征图谱,标定了11个共有特征峰,经与对照品比较,指认出6个,分别为迷迭香酸、丹酚酸B、二氢丹参酮Ⅰ、隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA,这6种成分在各自范围内线性关系良好,回收率在95.4%~99.9%之间,RSD均<3.0%。结论该方法操作简便、选择性高、稳定性好、分析速度快,可有效控制丹参药材的质量。Objective To establish the HPLC specific chromatogram of Salvia Miltiorrhiza from different producing places,and to simultaneously determine six active components.Methods The effective components were separated by HPLC with Capcell C18(4.6×250 mm,5 μm) in a gradient elution of acetonitrile and water with 0.05% H_3PO_4 as the mobile phase at the flow rate of 0.8 m L.min-1 and temperature of 30 ℃ and detection wavelength of 270 nm.Results The specific chromatogram of Salvia Miltiorrhiza was established and 11 common peaks were indicated. To compare to the standard,the six components was identified,rosmarinic acid,salvianolic acid B,dihydrotanshioneⅠ,crytotanshinone,tanshinoneⅠ,tanshinone ⅡA respectively. The six chemical components were well separated and with good liner relationship,the recoveries were between 95.4% and 99.9%,and the RSD was less than 3.0%.Conclusion The method is simple,accurate and repeatable,and can be used to control the quality of Salvia Miltiorrhiza.
