摘  要:目的探讨替普瑞酮(GGA)对糖尿病肾病(DN)大鼠Smad3和纤维连接蛋白(FN)表达的影响。方法将36只大鼠随机分为对照组,DN模型组,和干预组,单侧肾切除+腹腔注射链脲佐菌素建立DN大鼠模型,干预组每日给予GGA灌胃,免疫组化染色检测肾组织HSP72,Smad3和FN的表达。结果 GGA灌胃12周后,干预组肾组织免疫组化染色可检测到HSP72的表达;与对照组相比,DN模型组Smad3、FN的表达显著升高,而干预组Smad3、FN的表达较DN模型组降低,差异有统计学意义(P<0.05)。结论 GGA灌胃可诱导大鼠肾组织表达HSP72,并可降低DN大鼠的Smad3及FN的表达。Objective To determine whether geranylgeranylacetone affects on the protein expression of Smad3 and fibronectin in rat with diabetic nephropathy. Methods The thirty six rats were randomly divided into normal control group,diabetic nephropathy group and intervention group. The left nephrectomy and streptozotocin abdominal injection was chosen to establish the rat model of diabetic nephropathy. Geranylgeranylacetone was given by gavage each day for 12 weeks for the rats of intervention group. The expression of HSP72,Smad3 and fibronectin in kidney tissue was detected by immunohistochemistry. Results When geranylgeranylacetone had been gavaged for 12 weeks,the expression of HSP72 was detected by immunohistochemical staining in the intervention group. Compared with the diabetic nephropathy group,the intervention group had reduced protein levels of Smad3 and fibronectin. The difference is statistically significant. Conclusion The gavage of geranylgeranylacetone can induce the expression of HSP72 in rat kidney,and can significantly reduce the protein expression of Smad3 and fibronectin in rats with diabetic nephropathy.
