摘 要:目的:探索美洲大蠊促创面愈合相关活性物质。方法:采用多种色谱法分离纯化其非肽类小分子化合物,并借助谱学法鉴定其化学结构。结果:从中分离得到16个化合物,分别为2-(1,2,3-三羟基丙基)吡嗪(1)、2-甲基-6-(2,3,4-三羟基丁基)吡嗪(2)、3-甲基-6-(2,3,4-三羟基丁基)吡嗪(3)、3-羟基吡啶(4)、1-(2-羟乙基)-5-羟甲基-1H-吡咯-2-醛(5)、N-(5-乙酰吡咯-3-基)乙酰胺(6)、8-羟基-2,3-二氢-4(1H)-喹诺酮(7)、戊内酰胺(8)、N-乙酰多巴胺(9)、aspongopusin(10)、deoxyhyrtiosine A(11)、光色素(12)、6-羟甲基-3-羟基吡啶(13)、3,4-二羟基苯基甘油(14)、2-异戊基苯酚(15)和5-羟甲基呋喃-2-醛(16)。结论:化合物1、3、5和6为新的天然产物,化合物2、4、7、10~16为首次从美洲大蠊中分离得到;生物活性研究表明化合物12~14能促进人皮肤成纤维细胞迁移,尤以化合物13作用最为显著,而化合物3可促进人脐静脉内皮细胞血管生成。Objective: To explore compounds from Periplaneta americana and investigate their wound-healing promo- ting activities. Methods : Small molecules were isolated by using column chromatography and their structures were identified by spectroscopic methods. Results: Sixteen compounds isolated from P. americana were identified as 2-( 1, 2, 3-trihydroxyprop- yl) pyrazine ( 1 ), 2-methyl-6- (2, 3, 4-trihydroxybutyl) pyrazine (2), 3-methyl-6- ( 2, 3, 4-trihydroxybutyl) pyrazine ( 3 ), 3 -hydroxypyridine ( 4 ) , 1 - ( 2-hydroxyethyl ) -5 - ( hydroxymethyl ) -1H-pyrrole-2-carbaldehyde ( 5 ) , N- ( 5 -acetylpyrrol-3 -yl ) ac- etamide ( 6 ), 8-hydroxy-2, 3-dihydro4 (1H) -quinolone ( 7 ), valerolactam ( 8 ), N-acetyldopamine ( 9 ), aspongopusin ( 10), deoxyhyrtiosine A ( 11 ), lumichrome ( 12), 6-hydroxymethyl-3-pyridinol ( 13 ), 3, 4-dihydroxyphenylglycol ( 14 ), 2-(isopentyl) phenol ( 15 ), and 5-( hydroxymethyl ) furan-2-carbaldehyde ( 16 ), respectively. Conclusion: Compounds 1, 3, 5, and 6 are new naturally occurring compounds, and compounds 2, 4, 7, 10-16 were isolated from the title insect spe- cies for the first time. Biological evaluation revealed that compounds 12-14 could significantly stimulate HDFs migration with 13 to be the most active. In addition, compound 3 was found to be active towards HUVECs angiogenesis.
