摘  要:为探讨Jab1基因在乳腺癌细胞中的生物学功能,进一步揭示其作用的分子机制.首先在乳腺癌细胞MCF-7中,成功建立了慢病毒介导的Jab1基因的RNAi系统,并通过CCK8细胞增殖实验和细胞划痕实验证实Jab1基因表达的干扰使得MCF-7细胞的增殖和迁移发生显著的下降,细胞周期分析表明,干扰Jab1的表达能增加MCF-7细胞sub G1期的比例,诱导凋亡;此外,通过RT-PCR和Western blot实验首次发现Jab1能调控TBHS1的表达,添加甲基化转移酶抑制剂5-Aza-dC能抑制Jab1表达干扰介导的THBS1表达下调,提示Jab1可通过甲基化的表观遗传学途径调控THBS1基因的表达.In breast cancer cell line MCF-7,successfully established a lentivirus mediated RNAi of Jab1 gene expression.CCK8 cell proliferation assay and cell scratch experiments confirmed that knock down of Jab1 gene expression makes the proliferation and migration of MCF-7 cells were significantly decreased.Cell cycle analysis showed that knock down of Jab1 gene expression can increase the proportion of sub G1 phase in the MCF-7 cells and induce apoptosis.in addition,the RT-PCR and Western blot confirmed that Jab1 can regulate the expression of TBHS1,Add the methyltransferase inhibitor 5-Aza-dC can inhibit the expression of Jab1 interference mediated down-regulation of THBS1 expression,suggesting that Jab1 can regulate the expression of THBS1 by epigenetic signaling pathways.This research will be helpful to reveal the molecular mechanism of Jab1-THBS1 signaling pathway in Breast cancer cells.
