摘  要:目的探讨CD34^+造血干细胞移植在大鼠缺氧缺血性脑病中的神经保护作用和机制。方法建立体外人造血干细胞(CD34^+)增殖模型并进行体外扩增,分析其生物学活性。选取90只SD大鼠构建大鼠缺氧缺血动物模型并随机分为三组(每组30只):对照组、模型组和治疗组;给予治疗组大鼠造血干细胞(CD34^+)移植3 d后,处死大鼠并观察脑组织病理改变;采用TUNEL法检测神经细胞凋亡的发生;采用免疫组织化学法检测脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)和血清神经生长因子(nerve growth factor,NGF)蛋白的表达。结果与对照组相比,模型组和治疗组动物行为学异常明显加重,凋亡细胞数明显增多,细胞内BDNF和NGF含量显著下降(P<0.05);与模型组相比,治疗组动物行为学异常明显减轻,凋亡细胞数明显减少,细胞内BDNF和NGF含量显著上升(P<0.05)。结论 CD34^+造血干细胞移植对大鼠缺氧缺血性脑病中具有显著的神经保护作用,可以有效促进脑内神经营养相关因子的表达。Objective To investigate neuroprotective effect and mechanism of CD34^+hematopoietic stem cell transplantation in rats with hypoxic-ischemic encephalopathy(HIE). Methods Human hematopoietic stem cell(CD34^+) proliferation model was established and amplified in vitro. The biological activity was analyzed. 90 SD rats were selected to establish the animal model of rats with HIE and randomized into three groups(30 rats each): control group, model group and treated group. Rats in treated group were sacrificed three days after the treatment of hematopoietic stem cell(CD34^+) transplantation. The pathological changes in brain tissue were observed. TUNEL was used to detect the occurrence of apoptosis in nerve cells. The expression of brain-derived neurotrophic factor(BDNF) and nerve growth factor(NGF) was assessed by immunohistochemistry. Results Compared with control group, behavioral abnormalities of rats in model and treated group were remarkable. The number of apoptotic cells increased and the contents of BDNF and NGF decreased significantly(P〈0.05). Compared with model group, behavioral abnormalities of rats in treated group were less.The number of apoptotic cells decreased and the contents of BDNF and NGF increased significantly(P〈0.05). Conclusion The neuroprotective effect of CD34^+hematopoietic stem cell transplantation in rats with HIE was remarkable.CD34^+hematopoietic stem cell transplantation could promote the expression of relevant neurotrophic factors in the brain effectively.
