摘 要:目的观察小鼠在不同生理阶段雌激素及其受体及白细胞介素13(IL-13)和肿瘤坏死因子α(TNF-α)含量的变化,探讨雌激素水平对小鼠免疫功能的影响。方法清洁级健康雌性C57BL/6小鼠60只,其中2月龄小鼠(40只)随机分为发情间期组(对照组)、妊娠组,10月龄小鼠拟为自然绝经组,每组20只。3个月后进行实验,RT-PCR技术检测3组小鼠脾脏IL-13、TNF-αmRNA、ERαmRNA和ERβmRNA水平,ELISA检测血清雌二醇(E_2)水平,比较三组间各个指标差异。结果与对照组相比,妊娠组血E_2水平[(82.27±8.70)pmol/L vs.(55.23±5.11)pmol/L]和ERαmRNA水平[(3.93±1.54)vs.(2.70±1.17)]显著升高,自然绝经组E_2水平[(23.52±4.46)pmol/L vs.(55.23±5.11)pmol/L]和ERαmRNA水平[(1.15±0.71)vs.(2.70±1.17)]显著降低(P<0.05);妊娠组小鼠脾脏IL-13、TNF-αmRNA水平显著降低,而绝经期小鼠脾脏IL-13、TNF-αmRNA水平显著升高,差异有统计学意义(P<0.05);三组小鼠ERβmRNA水平无显著性差异(P>0.05)。结论绝经组小鼠IL-13、TNF-α浓度水平较妊娠组及发情间期组升高,可能与其雌激素水平及ERα受体下降有关。Objective:To observe estrogen receptor level and the content change of interleukins 13(IL-13)and tumor necrosis factor alpha(TNF-α)in different physiological stages of change in mice,and explore the effect of estrogen on immunity.Methods:Sixty clean grade healthy female C57 BL/6 mice were selected.Among them,40 mice with two month old were randomly divided into estrus group(control group)and pregnancy group,and 20 mice with ten month old were proposed to be the natural menopause group.PT-PCR technique was used to detect spleen IL-13,TNF-α,ERαand ERβmRNA level,and ELISA was used to detect serum E_2 level.The differences of the indicators were compared among the three groups.Results:Compared with control group,the serum E_2 levels[(82.27±8.70 pmol/L vs.55.23±5.11 pmol/L)]and ERαmRNA level[(3.93±1.54)vs.(2.70±1.17)]were significantly increased in pregnancy mice(P〈0.05).The E_2 level[(23.52±4.46)pmol/L vs.(55.23±5.11)pmol/L]and ERαmRNA level[(1.15±0.71)vs.(2.70±1.17)]were significantly decreased in natural menopause group(P〈0.05).The IL-13 and TNF-αmRNA levels in spleen of the pregnant mice were significantly decreased,while the IL-13 and TNF-αlevels in spleen of the menopausal mice were significantly increased(P〈0.05).There was nosignificant difference in ERβmRNA level among the three groups(P〉0.05).Conclusions:IL-13 and TNF-αexpression levels in postmenopausal mice were significantly increased compared with the estrus mice,which may be associated with decreased levels of estrogen and ER alpha receptors in postmenopausal mice.
