摘 要:目的探讨来曲唑诱导的多囊卵巢综合征(PCOS)大鼠血清与卵巢组织中内皮抑素(ES)、内皮素-1(ET-1)和血小板反应素-1(TSP-1)的表达及意义,并对比给予生长激素前后ES、ET-1和TSP-1的表达变化。方法选取6周龄清洁雌性SD大鼠70只,分为对照组20只,模型组50只。模型组给予来曲唑造模,剂量1mg/(kg·d),连续灌胃21d建立PCOS大鼠模型,对照组给予等剂量1%羧甲基纤维素灌胃。随机选取模型组10只大鼠评估造模成功后,模型组余下大鼠随机分为生长激素组(GH组,20只)和PCOS组(20只)。GH组皮下注射重组人生长激素(rhGH),剂量为1U/(kg·d),PCOS组和对照组皮下注射与rhGH等量的生理盐水,均21d后处死,留取各组大鼠血清样本及卵巢组织。采用ELISA法、免疫组织化学法检测各组大鼠血清、卵巢组织中ES、ET-1和TSP-1的表达。结果与对照组相比,PCOS组大鼠血清及卵巢组织中ES和ET-1均表达增加(P<0.05),TSP-1表达降低(P<0.05);与PCOS组大鼠相比,GH组大鼠血清及卵巢组织中ES和ET-1表达降低(P<0.05),而TSP-1表达增加(P<0.05)。结论 ES、ET-1及TSP-1在PCOS的发生发展中发挥着重要作用,GH可能通过调节血清及卵巢组织中ES和ET-1的表达而在PCOS中发挥作用。Objective:To explore the expression and significance of endostatin(ES),endothelin-1(ET-1)and thrombocytopenin-1(TSP-1)in the serum and ovary tissues of rats with polycystic ovary syndrome(PCOS)induced by letrozole,and compare the changes of ES,ET-1 and TSP-1 before and after administration of growth hormone.Methods:Seventy female SD rats of six weeks age were randomly divided into control group(n=20)and model group(n=50).In model group,the rate were administered letrozole by intragastric infusion with a dose of 1 mg/(kg·d)for 21 days to create PCOS model.The control group was given equal dose of 1% carboxymethyl cellulose.Ten rats were randomly selected to evaluate the success of establishment of model in model group,and the rest of rats were randomly divided into growth hormone group(GH group,n=20)and PCOS group(n=20).The GH group was injected subcutaneously with recombinant human growth hormone(rhGH)at a dose of 1 U/(kg.d),and the rats in PCOS group and the control group were injected with equal dose of normal saline.All the rats were killed after 21 days,the serum samples and ovarian tissue were collected.The expression of ES,ET-1 and TSP-1 in serum and ovarian tissues of rats were detected by ELISA and immunohistochemistry.Results:Compared with control group,the expressions of ES and ET-1 were significantly higher in serum and ovary tissues of PCOS rats(P〈0.05)and expression of TSP-1 was significantly lower(P〈0.05).Compared with group PCOS rats,the expressions of ES and ET-1 in serum and ovary tissue of GH group were significantly decreased(P〈0.05),while the expression of TSP-1 was significantly increased(P〈0.05).Conclusions:ES,ET-1 and TSP-1 play important roles in the development of PCOS.GH may play a role in PCOS by regulating the expression of ES and ET-1 in serum and ovarian tissue.
