摘  要:目的探讨黑色素纳米颗粒(MNP)经聚赖氨酸(PLL)改性连接小干扰RNA(si RNA)后的细胞毒性,为其在基因治疗中的应用提供依据。方法制备新型水溶性MNP并经PLL螯合改性,依靠静电作用连接si RNA,分别与4T1乳腺癌细胞和Hep2喉癌细胞进行孵育,采用CCK8检测法观察细胞生长状况,并对材料的细胞毒性进行分级。结果 4T1乳腺癌细胞和Hep2喉癌细胞在1 000μg/ml及其以下的实验分组中均生长良好,形态饱满。CCK8检测法数据表明细胞生存率均大于80%,毒性分级达合格。结论 MNP经PLL改性连接si RNA所形成的复合物是一种生物安全性高,毒性小的新型纳米基因载体。Objective To investigate the cytotoxicity of melanin nanoparticles(MNPs) modified by poly-L-lysine(PLL), and to provide a basis for its use in gene therapy. Methods The novel water-soluble MNPs were prepared and modified by PLL, which were connected with siRNA by electrostatic interaction and incubated with 4 T1 breast cancer cells and Hep2 laryngeal cancer cells, respectively. The CCK8 assay was used to determine the cell growth, and the cytotoxicity of the materials was classified. Results The 4 T1 breast cancer cells and Hep2 laryngeal cancer cells grew well with full morphology in the study subgroups of 1 000 μg/μl and below. The data of CCK8 test suggested that the cell survival rate was greater than 80%, and the toxic grade was qualified. Conclusion The complex of MNPs modified by PLL and connected with siRNA is a novel nano-gene vector with high biosafety and low toxicity.
