摘　　要：以丙二酸二乙酯和硫脲为原料,经缩合,硫酸二甲酯甲基化两步反应合成了2-甲硫基-3-甲基-6-甲氧基嘧啶-4-酮。该路线使用硫酸二甲酯替换文献路线中的碘甲烷,并且将分步甲基化过程简化为一步甲基化,路线总收率达79.31%,提高了30%。通过熔点、MS、~1H NMR确证目标化合物的结构。该路线原料易得,收率高,成本低,操作过程简便,具有放大前景。2-Methylthio-3-methyl-6-methoxypyrimidine-4-one was synthesized through two reactions including condensation and methylation using dimethyl malonate and thiourea as starting materials. The total yield was up to 79.31%. The structures of target compound were confirmed using MS and ^1H NMR. The advantages of this method included commercialized raw materials, high yield, low cost, simple process, and easy industrialization.
