摘  要:目的采用多电极皮层脑电监测技术,通过局部给药的方式,观察白细胞介素-6(IL-6)对大鼠皮质发育障碍(MCD)痫性放电的影响。方法建立液氮皮层冰冻MCD模型:对新出生的SD大鼠进行脑皮质板局部冰冻损伤。模型建立后通过形态学、行为学等进行评价。手术前1 w对MCD模型小鼠进行皮层脑电埋置。术后1 w所有实验大鼠随机分成匹罗卡品组、匹罗卡品加IL-6组。模型诱导组大鼠氯化锂腹腔注射(180 mg/kg),20 h后给予硫酸阿托品1 mg/kg腹腔注射,30 min后再给予20 mg/kg匹罗卡品腹腔注射。模型诱导加IL-6组增加IL-6局部注射。注射前先进行30 min的基线监测,注射后直接进行电生理检测。监测完毕对动物行为学以及相关如潜伏期、持续时间等电生理指标进行分析。结果所有实验组大鼠注射匹鲁卡品后随即出现眨眼、节律性咀嚼、节律性点头,约30 min后出现Ⅳ级以上发作。但是IL-6注射组动作明显减轻,出现时间较晚。电生理结果显示,与模型诱导组相比(25.93±1.33)min,模型诱导加IL-6注射组表现为潜伏期明显延长(31.05±2.35)min,两组潜伏期发作时间有明显差异(P<0.01);与模型诱导组相比[(34.58±1.02)s],模型诱导加IL-6注射组表现为放电持续时间明显减少[(17.75±1.94)s],两组时间有明显差异(P<0.01);与模型诱导组相比[(28.83±4.26)次],模型诱导加IL-6注射组表现为自发性癫痫放电次数明显减少[(12.00±3.03)次],两组时间有明显差异(P<0.01)。结论 IL-6明显减少痫性放电阈值,减少放电持续时间,可能对癫痫发作有一定的保护作用。Objective The effects of interleukin-6(IL-6) on the epileptic discharges of malformations of cortical development(MCD) in rats were studied by the topical delivery of drugs and the monitoring technique of multi-electrode cortical electroencephalography( EEG). Methods MCD model was established by freezing the cortex with liquid nitrogen: new-born SD rats were subjected to local cryopreservation of cerebral cortex. After the establishment of the model,the morphology and behaviors were evaluated. One week before surgery,the MCD model mice were subjected to cortical EEG. All the rats were randomly divided into pilocarpine group and pilocarpine plus IL-6 group. Rats in the model group were injected intraperitoneally with lithium chloride(180 mg/kg),20 h later the rats were injected intraperitoneally with atropine sulfate 1 mg/kg,and 20 mg/kg pilocarpine after 30 min. The model plus IL-6 group used the local injection of IL-6. Baseline monitoring was performed 30 min prior to injection and electrophysiological testing was performed immediately after the injection. After monitoring,the behavior of the animal and other related electrophysiological indicators,such as latency,duration and so on were analyzed. Results All rats in the experimental group showed blinking,rhythmic chewing and rhythmic nodding immediately after injection of pilocarpine,and they had the episode of more than Class Ⅳ after 30 min. But the action of IL-6 injection group was significantly reduced and appeared later. The electrophysiological results showed that the incubation period was significantly prolonged(31. 05 ± 2. 35) min in the model-induced plus IL-6 injection group compared with the model induced group(25. 93 ± 1. 33) min. Compared with the model-induced group(34. 58 ± 1. 02) s,the latency period in the model-induced(17. 75 ± 1. 94) s was decreased significantly,and there was a significant difference between the two groups(P〈0. 01).
