摘 要:为了探究芽孢杆菌Bacillus atrophaeus B44和Bacillus subtilis S37在棉花上的定殖能力与防病效果的关系,为更合理的施用芽孢杆菌提供理论指导,本研究通过Spizizen法将质粒PGF4412分别转化到2株芽孢杆菌中进行绿色荧光标记,研究不同浓度的抗性筛选平板对转化率的影响,将标记的菌株施用到棉苗根部观察其在根上的定殖量,同时将不同浓度的菌株接种到棉苗上,测定其对棉苗立枯病的防治效果。结果表明:当氨苄青霉素终浓度为15μg/mL时芽孢杆菌B.atrophaeus B44转化率最高,且假阳性较低,而B.subtilis S37的终浓度为20μg/mL。施用GFP标记的芽孢杆菌后,在荧光显微镜下观察发现,这2株菌仅在棉苗根表定殖,不能定殖于棉苗根内部。且初始接菌量越高,2株菌在土壤中及棉苗根表的定殖量越高。其中B44在最大初始接菌量时对棉苗立枯病的防效最好,S37在初始接菌量为1×10~8 CFU/g时,对棉苗立枯病的防效达到51.7%。B.atrophaeus B44和B.subtilis S37可在棉苗根表定殖,且定殖量与棉苗立枯病的防效密切相关,具有良好的应用前景。The purpose of this study was to provide theoretical support for more reasonable use of B44 andS37. The relationship of two strains colonization number and their control effect to cotton Rhizoctoniosis wereinvestigated in this study. The plasmid PGF4412 with GFP was transformed into two strains by modified Spizizen, and the optimalresistance plates for transformation rate was selected. The colonization site on cotton root was observed under fluorescencemicroscope and colonization number was count on plates. Their control effects of different colonization number on cottonRhizoctoniosis were investigated. The results showed that when the final concentration of ampicillin was 15 μg/mL, B44 strainhave higher transformation rate and lower false positive rate while S37strain need 20 μg/mL concentration. The two strains wereonly colonized on surface of cotton seedling roots observed with fluorescence microscope. The higher amount of bacteria was usedinitially, the more colonization of the two strains in the soil and surface of cotton seedling roots. Besides B44 had more highercontrol effect on cotton Rhizoctoniosis when the initial dosage was higher; S37 had the best control effect with 51.7% when itsinitial dosage was the highest about1×10^8CFU/g. The colonization amount of two Bacillus were closely correlated with theircontrol effect on cotton Rhizoctoniosis. They have a good prospect of application.
