摘  要:目的观察单侧迷走神经切断对小鼠咳嗽及相关神经肽蛋白表达水平的影响。方法将30只BALB/c小鼠随机分为对照组、左侧假手术组、右侧假手术组、左侧手术组及右侧手术组,每组各6只。在辣椒素刺激诱发咳嗽的小鼠模型上进行左侧或右侧迷走神经切断术或假手术,术后第8天统计各组小鼠咳嗽次数,检测所有小鼠肺组织中神经激肽B(neurokinin B,NKB)表达水平。结果左侧手术组小鼠术前、术后咳嗽次数分别为(8.50±2.17)次和(2.00±0.67)次,右侧手术组小鼠术前、术后咳嗽次数分别为(10.33±2.55)次和(3.67±0.34)次,组内手术前后小鼠咳嗽次数比较均有显著差异(P<0.05)。术后,左侧手术组和右侧手术组小鼠肺组织中NKB水平分别为(8.67±1.41)pg/ml和(9.09±0.99)pg/ml,两组小鼠NKB水平均显著低于对照组[(10.81±1.56)pg/ml]、左侧假手术组[(10.97±2.83)pg/ml]及右侧假手术组[(9.77±0.89)pg/ml](P<0.05)。小鼠术后咳嗽次数与NKB表达水平呈正相关(r=0.739,P=0.041)。结论迷走神经在辣椒素刺激诱发咳嗽小鼠中有重要作用,主要通过神经肽的变化发挥作用,单侧迷走神经切断后,NKB水平降低,小鼠咳嗽减少。Objective To observe the effect of unilateral vagotomy on cough and related neuropeptide protein expression in mice. Method 30 BALB/c mice were randomly divided into control group, left sham operation group, right sham operation group, left operation group and right operation group, each group had 6 mice. The left and right vagotomy or sham operation were performed in capsaicin stimulation induced cough model mice, the number of cough in mice was counted in eighth days. The expression level of neurokinin B(NKB) in lung tissue of all mice was detected. Result Before and after operation, the frequency of cough of left operation group were(8.50±2.17) times and(2.00±0.67) times, and the right operation group were(10.33±2.55) times and(3.67±0.34) times respectively, there were significant differences in cough frequency before and after surgery in both groups(P〈0.05). Left operation group of postoperative NKB level was(8.67±1.41) pg/ml, right operation group was(9.09±0.99) pg/ml, the level of NKB in the two groups was significantly lower than that of the control group [(10.81±1.56) pg/ml], left sham operation group [(10.97±2.83) pg/ml] and right sham operation group [(9.77±0.89) pg/ml](P〈0.05). The cough frequency in mice was positively correlated with NKB expression level(r = 0.739, P = 0.041). Conclusion Vagus nerve stimulation in mice induced by capsaicin plays an important role in cough, mainly through the changes of neuropeptides play a role in unilateral vagotomy, neuropeptide secretion decreased, resulting in reduced cough.
