摘  要:目的:探索老年雄性SD大鼠前列腺导管内是否存在尿路上皮细胞及其形态特点。方法:取5只老年雄性SD大鼠,水合氯醛麻醉后取尿道、前列腺及膀胱组织,完成病理切片和HE染色,观察前列腺导管内尿路上皮细胞的表达情况。免疫组化检测SD大鼠3种组织内CK7、CK20和尿斑蛋白Ⅲ(UroplakinⅢ,UPⅢ)的表达情况。另取10只老年雄性SD大鼠,同法将膀胱、前列腺腹侧叶和前列腺侧叶组织取出碎冰保存,随机分为实验组和空白对照组,每组5只大鼠,以UPⅢ阳性设门检测不同部位前列腺组织中是否存在UPⅢ阳性的细胞及其表达情况。结果:HE染色可观察到SD大鼠前列腺导管开口处存在形态与膀胱和尿道尿路上皮细胞相似的尿路上皮细胞,但该细胞未呈现典型的伞状细胞结构;其免疫组化CK7和UPⅢ阳性、CK20阴性。随着前列腺导管延伸至管腔底部,CK7和UPⅢ的表达逐渐减低,直至消失。流式细胞学检测结果表明前列腺腹侧叶及侧叶组织内有UPⅢ阳性的细胞,该细胞分别占各组织内细胞总数的(55.96±3.85)%和(50.76±3.99)%。结论:老年雄性SD大鼠前列腺导管内存在与膀胱尿路上皮相似的尿路上皮细胞,但该细胞未呈现典型的伞状细胞形态结构;该细胞免疫组化CK7和UPⅢ表达阳性。Objective: To investigate the presence of urothelial cells in the prostate ducts of aged male SD rats and their morphological characteristics. Methods: 5 aged male SD rats were anesthetized with chloral hydrate for urethral,prostate and bladder tissues.Pathological sections and HE staining were performed to observe the expression of prostate duct urothelial cells. Immunohistochemical method was used to detect the expression of CK7,CK20 and Uroplakin III( UP III) in three tissues of aged SD rats. Another 10 aged male SD rats used the same method for bladder,ventral lobe prostate and lateral lobe prostate tissue,used ice to storage,these rats were randomly divided into experimental group and control group,5 rats in each group,with UP III positive set of prostate tissue detection in different parts of prostate,and the existence of cells the positive expression of UP III. Results: HE staining could be observed in the aged SD rat prostate duct urothelial cell morphology and was similar to the bladder and urethra urothelial cells,but the cell did not show the typical structure of umbrella cells,the immunohistochemistry of CK7,UP III positive and CK20 negative. The expression of CK7 and UP III decreased gradually with the development of the prostatic duct extending to the bottom of the lumen. The results of flow cytometry showed that there were UP III positive cells in the tissues of bladder,ventral lobe and lateral lobe of the prostate,which accounted for55. 96 ± 3. 85% and 50. 76 ± 3. 99% of the total cells in the prostate tissues,respectively. Conclusion: There are urothelial cells similar to the urothelium of the urinany tract in the prostate duct of the aged male SD rats,but the cells don't exhibit typical umbrella cell morphology and the cells are positive for immunohistochemical CK7 and UPIII expression.
