摘  要:目的:研究测定愈疡散中主要成分紫丁香苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1、长梗冬青苷和黄芪甲苷的含量,同时建立其皂苷类成分的HPLC指纹图谱分析方法,为科学评价与有效控制愈疡散的质量提供依据。方法:采用高效液相色谱-蒸发光散射检测法(HPLC-ELSD法),使用C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-水为流动相,梯度洗脱,流速1.0 m L·min^(-1),柱温35℃,检测器参数为蒸发温度为100℃,气体流量为2.8 L·min-1,增益值为1.0。结果:紫丁香苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb_1、长梗冬青苷和黄芪甲苷6个成分的含量范围分别为0.662~2.638、0.840~1.358、7.305~11.704、3.429~6.343、3.048~4.248、1.375~3.841 mg·g^(-1),各项方法学考察结果表明该法符合含量测定的要求;同时10批次愈疡散样品得到的指纹图谱相似度均大于0.96,标定共有峰11个,并对其中7个色谱峰进行了归属。结论:所建立的指纹图谱特征性强;所建立的指纹图谱结合主要功效成分多指标含量测定方法能更加客观有效地控制壮药愈疡散制剂的质量。Objective:To determine the content of syringin,notoginsenoside R1,ginsenoside Rg1,ginsenoside Rb1,pedunculoside and astragaloside Ⅳ and to establish an HPLC fingerprint of saponins in Yuyangsan,so as to provide a scientific evaluation and effective control for the quality control.Methods:HPLC-ELSD method was used.The analysis was carried on a C18 column(250 mm×4.6 mm,5 μm)with acetonitrile-water as the mobile phase in gradient elution at a flow rate of 1 mL·min^-1.The column temperature was 35 ℃ and the drift tube temperature was 100 ℃.The gas flow rate was 2.8 L·min^-1 and the gain was 1.0.Results:The content ranges of syringin,notoginsenoside R1,ginsenoside Rg1,ginsenoside Rb1,pedunculoside and astragaloside Ⅳ were 0.662-2.638 mg·g^-1,0.840-1.358 mg·g^-1,7.305-11.704 mg·g^-1,3.429-6.343 mg·g^-1,3.048-4.248 mg·g^-1 and 1.375-3.841 mg·g^-1,respectively.And the methodological study results showed that the method met the requirements of determination.Similarities of 10 batches of samples were all up to 0.96 with 11 common peaks,7 of which were identified.Conclusion:The developed fingerprint is specific.The established method combining fingerprint with multi-index assay can control the quality Yuyangsan in a more objective and effective way.
