摘  要:目的:建立高效液相色谱法同时测定软肝缩脾丸中芍药苷、丹酚酸B、大黄酸、五味子醇甲、大黄素、大黄酚、丹参酮ⅡA 7个有效成分的含量。方法:采用Wondasil C18色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,流速1.0 m L·min-1,柱温30℃,检测波长230 nm(0~34.0 min,检测芍药苷、丹酚酸B、大黄酸)、217 nm(34.0~37.0 min,检测五味子醇甲)、225 nm(37.0~56.0 min,检测大黄素、大黄酚)、270 nm(56.0~60.0 min,检测丹参酮ⅡA)。结果:芍药苷、丹酚酸B、大黄酸、五味子醇甲、大黄素、大黄酚、丹参酮ⅡA的线性范围分别为10.04~250.95μg·mL^(-1)(r=0.999 5)、10.25~256.28μg·mL^(-1)(r=0.999 7)、0.84~21.07μg·mL^(-1)(r=0.999 5)、0.91~22.86μg·mL^(-1)(r=0.999 2)、0.67~16.72μg·mL^(-1)(r=0.999 1)、1.45~36.30μg·mL^(-1)(r=0.999 5)、0.38~9.40μg·mL^(-1)(r=0.999 3);平均加样回收率(n=9)为99.5%~101.6%,RSD为0.61%~1.1%。样品中上述7个有效成分的含量范围分别为2.327~2.835、2.423~3.158、0.051~0.207、0.052~0.326、0.089~0.155、0.330~0.481、0.045~0.087 mg·g-1。结论:本法可用于软肝缩脾丸中芍药苷、丹酚酸B、大黄酸、五味子醇甲、大黄素、大黄酚、丹参酮ⅡA 7个有效成分的含量测定。Objective:To establish an HPLC method for simultaneous determination of paeoniflorin,salvianolic acid B,rhein,schisandrin,emodin,chrysophanol,and tanshinone ⅡA in Ruangan Suopi pills.Methods:The chromatographic separation was performed on a Wondasil C18 column(250 mm×4.6 mm,5 μm)at 30 ℃ with acetonitrile(A)-0.1% phosphoric acid solution(B)as mobile phase in a gradient elution.The flow rate was 1.0 mL·min^-1 and the UV detection wavelength was set at 230 nm(0-34.0 min for paeoniflorin,salvianolic acid B and rhein),at 217 nm(34.0-37.0 min for schisandrin),at 225 nm(37.0-56.0 min for emodin and chrysophanol),and at 270 nm(56.0-60.0 min for tanshinone ⅡA).Results:The linear ranges of paeoniflorin,salvianolic acid B,rhein,schisandrin,emodin,chrysophanol,and tanshinone ⅡA were 10.04-250.95 μg·mL^-1(r=0.999 5),10.25-256.28 μg·mL^-1(r= 0.999 7),0.84-21.07 μg·mL^-1(r=0.999 5),0.91-22.86 μg·mL^-1(r=0.999 2),0.67-16.72 μg·mL^-1(r=0.999 1),1.45-36.30 μg·mL^-1(r=0.999 5),0.38-9.40 μg·mL^-1(r=0.999 3),respectively.The average recoveries(n=9)were 99.5%-101.6% with RSD of 0.61%-1.1%.The contents of above-mentioned seven constituents in samples were 2.327-2.835 mg·g^-1,2.423-3.158 mg·g^-1,0.051-0.207 mg·g^-1,0.052-0.326 mg·g-1,0.089-0.155 mg·g^-1,0.330-0.481 mg·g^-1,0.045-0.087 mg·g^-1,respectively.Conclusion:The method can be used to determine the content of paeoniflorin,salvianolic acid B,rhein,schisandrin,emodin,chrysophanol,and tanshinone ⅡA in Ruangan Suopi pills.
