摘 要:目的检测儿童类风湿关节炎亚型(S-JIA、JIA少关节型)外周血Nod样受体家族包含pyrin结构域蛋白-3(NLRP-3) mRNA基因表达差异,探讨NLRP-3 mRNA表达水平差异与患儿临床表现特征的相关性。 方法收集28例初治类风湿关节炎(S-JIA13例,JIA少关节型15例)住院患儿及20例同期正常体检患儿的外周血标本,提取标本总RNA,实时定量反转录聚合酶链反应(RT-qPCR)检测外周血标本中NLRP-3 mRNA表达的差异。 结果在外周血中NLRP-3 mRNA在S-JIA组的相对表达量为0.066 7±0.004 4,高于JIA少关节型组(0.010 7±0.001 5)及正常对照组(0.021 3±0.003 2),差异有统计学意义(F=5.491,P=0.010);正常对照组与JIA少关节型组之间比较时,差异无统计学意义(F=1.745,P=0.105)。 结论NLRP-3 mRNA水平升高,是导致S-JIA产生炎性反应的重要影响因素之一。ObjectiveTo examine the gene expression difference of NLR family, pyrin domain containing-3 (NLRP-3) mRNA in peripheral blood of the various subtypes of juvenile rheumatold arthritis (S-JIA and oligoarticular JIA), and analyze the correlation between the expression level of NLRP-3 mRNA and the clinical features of children. MethodsThe peripheral blood from 28 cases of juvenile rheumatold arthritis (including 13 cases of S-JIA13 and 15 cases of oligoarticular JIA) admitted and 20 healthy children receiving the healthy examination at the same time in the First Affiliated Hospital of Zhengzhou University from May 2014 to August 2015 were investigated. The total RNA was extracted, and the expression difference of NLRP-3 mRNA in peripheral blood was detected by real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR). ResultsIn the peripheral blood, the relative expression of NLRP-3 mRNA in the S-JIA group was (0.066 7±0.004 4), higher than that in oligoarticular JIA group (0.010 7±0.001 5) and normal control group (0.021 3±0.003 2) with the difference being statistically significant (F=5.491, P=0.01). There was no statistically significant difference between the normal control group and the oligoarticular JIA group (F=1.745, P=0.105). ConclusionThe increased level of NLRP-3 mRNA is one of the most crucial factors to lead to S-JIA to produce inflammatory response.
