松郁安神方对异相睡眠剥夺大鼠海马CREB表达的影响

摘　　要：目的观察松郁安神方对异相睡眠剥夺(PSD)大鼠海马环磷酸腺苷反应元件结合蛋白(CREB)的影响。方法采用改良多平台睡眠剥夺法建立PSD大鼠模型,连续7 d用松郁安神方进行灌胃。RT-PCR和蛋白印迹法检测对照组、PSD模型组、中药高、低剂量组大鼠海马CREB m RNA和蛋白表达的情况。结果与对照组比较,PSD模型组大鼠海马CREB m RNA及蛋白表达均明显降低(P<0.01),不同剂量松郁安神方干预后,中药高剂量组CREB m RNA及蛋白表达明显升高(P<0.01),中药低剂量组CREB m RNA表达升高(P<0.05),蛋白表达也明显升高(P<0.01)。结论松郁安神方改善PSD大鼠学习记忆能力的机制可能与调节海马CREB的表达有关。Objective： To observe the effects of the Songyu Anshen Recipe on levels of CREB in the hippocampus of rats with paradoxical sleep deprivation （PSD）. Methods： The PSD model was established by modified multi-platform sleep deprivation method, the formula of Songyu Anshen Recipe was given by gavage for seven days. The expression of CREB mRNA and protein in hippocampus of rats was detected by RT-PCR and protein imprinting technique in control group, PSD model group, high- and low-dose group. Results： Compared with the control group, the expression of CREB mRNA and protein in hippocampus of the PSD model group decreased significantly （P〈 0.01）. After the intervention of different doses of Songyu Anshen Recipe, the expression of CREB mRNA and protein in the high-dose group increased significantly （P〈 0.01）, the expression of CREB mRNA in the low-dose group increased （P〈 0.05） and its protein expression also increased markedly （P〈 0.01）. Conelusion： The mechanism of Songyu Anshen Recipe on improving the learning and memory ability of PSD rats may be related to the regulation of CREB expression in hippocampus.
