肝病实脾法对急性肝损伤小鼠肝抗氧化能力的影响

摘　　要：目的观察健脾益气方四君子汤、常用治肝方四逆散防治急性肝损伤小鼠过氧化损伤情况,以探讨肝病实脾法对肝抗氧化能力的影响。方法将50只BALB/C雄性小鼠,随机分成对照组、模型组、四君子汤组、四逆散组、四君子汤合四逆散组5个组。造模前对治疗组给予灌胃给药,对照组、模型组给予蒸馏水灌胃,每天1次,共4 d。最后1次给药后4 h造模,进行尾静脉注射刀豆蛋白(Con A,2 mg/mL)0.2 mL/只,以诱导肝损伤,其中对照组均给予尾静脉注射PBS 0.2 mL/只。造模后第6 h取小鼠血清和肝组织,检测谷草转氨酶(AST)、谷丙转氨酶(ALT)、一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)水平。结果四逆散组、四君子汤合四逆散组均能降低AST、ALT、NO、MDA水平,与模型组比较有显著性意义(P<0.05);与四逆散组相比,四君子汤合四逆散组能更好降低AST、ALT、MDA、NO水平(P<0.05)。结论益气健脾的实脾法能在肝病治疗中通过提高肝抗氧化能力实现对肝组织的保护作用。Objective： To observe the effects of ＂Sijunzi Decoction＂ used for spleen-strengthening and qi-invigorating and ＂Sini San＂ commonly used for liver disease on peroxidative damage in mice with acute liver injury, so as to explore the effects of ＂reinforcing the spleen in treatment of liver disease＂ on liver＇s antioxidant ability. Methods： Fifty male BALB/C mice were randomly divided into 5 groups： control group, model group, Sijunzi Decoction group, Sini San group, Sijunzi Decoction combined with Sini San group. The treatment groups were given medicine respectively by gavage, while the control group and model group were given distilled water by gavage, once a day over 4 days. At the 4th hour after the last gavage, control group was given PBS 0.2 mL injected through caudal vein, while the other groups were given ConA 0.2 mL per mouse （2 mg/mL） injected through caudal vein to induce hepatic injury. Blood and liver tissues were collected at the 6 hour after modeling to detect the level of AST、ALT、NO 、MDA、SOD、GSH. Results： Compared with the model group, Sini San group and Sijunzi Decoction combined with Sini San group can significantly reduce the level ofAST, ALT, NO and MDA （P〈0.01）. Compared with Sini San group, Sijunzi Decoction combined with Sini San group can better reduce the level of AST, ALT, MDA and NO （P〈 0.01）. Conclusion： ＂Reinforcing the spleen＂ of spleen-strengthening and qi-invigorating can protect the liver tissues through improving the hepatic antioxidation ability in the treatment of liver diseases.
