摘  要:目的观察加味阳和汤抗硬皮病皮肤损伤的可能机制。方法雌性BALB/c小鼠40只,随机分为正常组、空白组各8只,造模组24只。正常组不注射,空白组予背部皮下注射生理盐水0.1 mL,造模组小鼠皮下注射浓度为500μg/mL的博莱霉素0.1 mL/d,连续4周,制备硬皮病模型。造模成功后随机分为模型组、阳和汤组、地塞米松组各8只。各组分别予生理盐水(空白组和模型组)、加味阳和汤(阳和汤组)、地塞米松(地塞米松组)灌胃,4周后取皮肤组织,行HE染色,PCR和Western blot分别检测小鼠皮肤β-Catenin的m RNA及蛋白表达量。结果治疗后,小鼠皮肤HE染色与正常组比较:空白组无明显差异;模型组真皮层增厚,排列紊乱,皮下脂肪层变薄;阳和汤组真皮层稍变薄,胶原纤维束排列较疏松,皮下脂肪层稍变薄。与正常组比较,模型组β-Catenin m RNA及蛋白表达水平上调,P-β-Catenin蛋白表达水平下调(P<0.05);与模型组比较,阳和汤组和地塞米松组β-Catenin m RNA及蛋白表达水平下调,P-β-Catenin蛋白表达水平上调(P<0.05)。结论加味阳和汤能降低硬皮病小鼠皮肤β-Catenin m RNA及蛋白表达,从而起到抗皮肤纤维化的治疗作用。Objective: To observe the possible mechanism of modified Yanghe Decoction (YHD) on the skin of the mouse model with systemic sclerosis. Methods: 40 female BALB/c mice were randomly divided into normal group (n=8), control group (n = 8) and model group (n=24). Except for the normal group, the control group and model group were injected subcutaneously with 0.1mL normal saline and 500 Ixg/mL bleomycin respectively over a period of 4 weeks to establish the scleroderma model. The 24 model mice were randomly allocated into the model group, the YHD group and the dexamethasone (DXM) group. The control group and the model group were fed with normal saline, the DXM group and YHD group were fed with DXM and YHD respectively. After 4 weeks, the mice were executed and skin samples were used for HE staining, while qPCR and western blot were used to detect the relative expression levels of mRNA and protein of β-Catenin. Results: After treatment, compared with the normal group, the control group showed no significant difference, while the model group indicated that the dermis was thicker, disordered and the subcutaneous fat layer became thinner. In the YHD group, the dermis was thinner, collagen fiber bundles were loosely arranged and the subcutaneous fat layer was a bit thinner. Compared with the normal group, the β-Catenin mRNA and the expression of protein in model group were increased, P-β-Catenin was decreased (P〈0.05). Compared with the model group, the β-Catenin mRNA and the expression of protein in YHD group and DXM group were decreased, P-β-Catenin was increased (P〈0.05). Conclusion: Modified Yanghe Decoction can reduce the skin β-Catenin mRNA and protein expression in scleroderma mice and plays a therapeutic effect against skin fibrosis.
