Research of Resveratrol on Alleviating Insulin Resistance and Regulating Liver Mitochondrial Function in Mice Fed with High Fat Diet

摘  要:目的研究白藜芦醇调节线粒体功能、改善高脂诱导的胰岛素抵抗的机制。方法 8周龄SPF级雄性C57BL/6J小鼠30只,随机分为低脂对照组(n=10)、高脂模型组(n=20),分别用低脂饲料(4.5%脂肪)、高脂饲料(45%脂肪)喂养。喂养12周后,通过葡萄糖耐量实验(IPGTT)检测胰岛素抵抗模型。模型成功后随机分为模型组(n=10)和药物组(n=10),药物组给予白藜芦醇灌胃干预,对照组和模型组给予生理盐水灌胃,连续干预10 d后进行IPGTT实验并用免疫蛋白印迹方法(Western Blot)检测肝脏组织的线粒体形态调节信号蛋白、炎症信号蛋白以及Ca^++Mg^++、Na^+K^+ATP-Pase含量。结果白藜芦醇可以促进小鼠肝脏组织中促线粒体融合蛋白Mfn2、OPA1表达,抑制线粒体促分裂蛋白Drp1蛋白含量,并且增加肝脏组织中Ca^++Mg^++、N^a+K^+ATP-pase含量。白藜芦醇可以明显增加小鼠肝脏组织中NF-k B的抑制性蛋白IκBα的表达,相应地减少肿瘤坏死因子(TNF-α)蛋白的表达。结论白藜芦醇短期灌胃干预可以通过线粒体形态与功能的调控以及相关的炎性信号变化改善高脂和肥胖等病理情况下的胰岛素敏感性。Objective: To study the mechanism ofresveratrol on regulating mitochondrial function and improving insulin resistance induced by high-fat diet. Methods: Thirty 8-week-old SPF male C57BL/6J mice were randomly divided into low-fat control group (n= 10) and high-fat model group (n =20). Both groups were fed with low-fat diet (4.5% fat) and high-fat diet (45% fat) respectively. After 12 weeks of feeding, the insulin resistance model was tested by glucose tolerance test (IPGTT). The successful models were randomly divided into two groups: the model group (n = 10) and the drug group (n = 10). The drug group was given resveratrol by gavage, while the control group and model group were given normal saline in the same way. After 10 days of gavage, an IPGTT experiment was performed and Western blot was also used to detect mitochondrial morphology signal proteins, inflammatory signaling proteins and the level of Ca^++, Mg^++, Na^+, K^+ ATP-Pase in liver tissue. Results: Resveratrol can promote the expression of mitochondrial fusion protein Mfn2 and OPAl in mouse liver tissue, suppress the content of mitochondrial mitogenic protein Drpl, and increase the content of Ca^++, Mg^++, Na^+, K^+ ATP-Pase in liver tissue. Resveratrol can significantly increase the expression of IKBα of NF-κB inhibitory protein in mouse liver tissue, and correspondingly reduce the expression of tumor necrosis factor (TNF-α). Conclusions: Short-term intragastric intervention of resveratrol can improve insulin sensitivity in pathological conditions such as high fat and obesity through mitochondrial morphology and function regulation and related inflammatory signaling changes.
